摘 要:目的观察电针对失神经兔骨骼肌中核转录因子-κB(NF-κB)、转化生长因子-β1(TGF-β1)表达的影响,探讨其对失神经骨骼肌萎缩的作用及可能分子机制。方法将30只新西兰兔随机分为空白对照组、模型组和电针组,每组10只。模型组和电针组采用钳夹胫神经的方法制备失神经骨骼肌萎缩兔模型。造模后第2天,电针组术侧下肢进行电针干预。造模14 d后测定各组兔腓肠肌湿重比、肌纤维截面积等。采用实时荧光定量聚合酶链式反应检测各组兔腓肠肌组织中NF-κB、TGF-β1 mRNA的表达,采用蛋白质印迹检测NF-κB、TGF-β1蛋白的表达。结果空白对照组、模型组和电针组新西兰兔的最终存活数分别为10、8、7只。与空白对照组比较,模型组和电针组兔腓肠肌湿重比、肌纤维截面积均明显下降,差异均有统计学意义(P<0.05);电针组兔腓肠肌湿重比、肌纤维截面积均明显高于模型组,差异均有统计学意义(P<0.05)。与空白对照组比较,模型组兔腓肠肌NF-κB、TGF-β1 mRNA表达水平均明显升高,差异均有统计学意义(P<0.05);电针组兔腓肠肌NF-κB、TGF-β1 mRNA及蛋白表达水平均较空白对照组明显升高,较模型组明显降低,差异均有统计学意义(P<0.05)。结论电针可通过下调腓肠肌NF-κB、TGF-β1的表达而改善骨骼肌萎缩程度,这可能是电针延缓失神经性骨骼肌萎缩的机制之一。Objective To observe the effects of electroacupuncture on the expression of nuclear factor-kappa B(NF-κB)and transforming growth factor-β1(TGF-β1)in the skeletal muscle of denervated rabbits,and to explore its effect on denervated skeletal muscle atrophy and its possible molecular mechanism.Methods Thirty New Zealand rabbits were randomly divided into the blank control group,model group and electroacupuncture group,10 cases in each group.The model group and electroacupuncture group adopted the clamping rabbit tibial nerve method to prepare the denervated skeletal muscle atrophy model.On 2nd after modeling,the electroacupuncture group performed the electroacupuncture intervention on the operative side lower limbs.After 14 d of modeling,the wet-weight ratio and muscle fiber cross-sectional of the gastrocnemius muscle in each group were measured,and the mRNA expressions of NF-κB and TGF-β1 in gastrocnemius muscle tissue were detected by adopting the real-time fluorescence quantitative PCR.Western blotting was used to measure the protein expression of NF-κB and TGF-β1.Results The final survival number of New Zealand rabbits were 10 cases in the blank control group,8 cases in the model group and 7 cases in the electroacupuncture group.Compared with the blank control group,the wet-weight ratio and muscle fiber cross-sectional area of gastrocnemius muscle in the model group and electroacupuncture group were significantly decreased,and the differences were statistically significant(P<0.05),while the wet-weight ratio and muscle fiber cross-sectional area in the electroacupuncture group were significantly higher than those in the model group,and the differences were statistically significant(P<0.05).Compared with the blank control group,the mRNA expression levels of NF-κB and TGF-β1 of the gastrocnemius increased in the model group,and the difference was statistically significant(P<0.05),while the mRNA and protein expression levels of NF-κB and TGF-β1 in the electroacupuncture group were significantly increased
