摘 要:含水量是影响种子超低温保存效果的关键因子,通过对不同含水量的冻存后种子相关生理生化指标的研究,从生理水平探讨含水量对冷冻保存效果的影响方式,揭示其冷冻/保护生理机制。结果表明,含水量对冻存后种子各项生理指标存在极显著的影响,含水量为6.1%时,MDA、REC含量最低;SOD,POD,CAT活性,AsA、可溶性糖(SS)、可溶性蛋白(SP)含量均最大,表明此含水量下细胞抗氧化能力强,膜系统损伤小,细胞代谢旺盛,超低温保存效果好,进一步从生理水平验证6.1%的含水量为马尾松种子超低温保存的最适含水量。Water content is the key factor affecting the cryopreservation effect of seeds.Based on the study of the related physiological and biochemical indexes of seeds with different water content after cryopreservation,the effects of water content on cryopreservation was discussed from the physiological level and the physiological mechanism of damage or protection was further revealed.The results showed that the content of malondialdehyde(MDA)content and relative conductivity(REC)was the lowest when the water content was 6.1%.The activities of superoxide dismutase(SOD),peroxidase(POD),catalase(CAT),ascorbic acid(AsA),soluble sugar(SS)and soluble protein(SP)were the highest,indicating that the cells had strong antioxidant capacity,less membrane system damage,vigorous cell metabolism,and good cryopreservation effect under such water content.It was further verified from physiological level that 6.1%water content was the optimal water content for the seed cryopreservation of Pinus massoniana seeds.
