金黄色葡萄球菌coa和saeRS基因对小鼠部分生物学指标的影响

摘  要:试验旨在探索金黄色葡萄球菌saeRS和coa对小鼠体重、感染部位皮内细菌数、感染部位伤疤面积及机体产生IL-17a的影响。利用金黄色葡萄球菌923及其突变菌株923/coa、923/saeRS和923/coa/saeRS皮内感染小鼠背部皮肤,测定感染后不同时间点各试验组小鼠体重、感染部位皮内细菌数、血清IL-17a浓度及感染部位的伤疤面积,计算小鼠血清IL-17a浓度与伤口面积之间的相关系数。结果表明:(1)923/coa/saeRS菌株感染组体重显著(P<0.05)/极显著(P<0.01)高于923/saeRS和923/coa感染组,923/saeRS感染组体重极显著(P<0.01)高于923/coa菌株感染组;(2)每克感染部位皮肤内细菌数,923菌株感染组显著(P<0.05)/极显著(P<0.01)高于923/coa、923/saeRS和923/coa/saeRS菌株感染组;(3)感染细菌后,小鼠血清IL-17a浓度逐渐升高,在24 h达到浓度峰值,随后逐渐下降;感染后8 h,菌株923组血清IL-17a浓度显著(P<0.05)低于923/saeRS组;(4)923/saeRS和923/coa/saeRS感染组小鼠皮肤伤疤面积显著(P<0.05)/极显著(P<0.01)低于923和923/coa感染组;(5)感染后24 h血清IL-17a浓度与感染后120 h小鼠感染部位伤疤面积的相关系数较强,并且达到极显著水平(P<0.01)。coa和saeRS基因缺失降低菌株923的毒性,使突变菌株感染小鼠体重提高,感染部位细菌含量降低,感染部位皮肤伤疤面积减小,血清IL-17a浓度降低。The purpose of this study was to investigate the effects of saeRS and coa on body weight,the number of bacteria in the infected area,the lesion size and the concentration of IL-17a in host serum.Wild type strain 923 and deletion mutants 923/coa,923/saeRS and 923/coa/saeRS were infected on the back of mice intradermally.Body weight,bacterial number in infected area,the lesion size and the concentration of IL-17a in host serum were measured at different experimental time,the correlation coefficient between serum IL-17a concentration and skin lesion was calculated.Results showed that:(1)Double deletion mutant 923/coa/saeRS enhanced host body weight significantly than single mutant 923/saeRS(P<0.05)and single mutant 923/saeRS,single mutant 923/saeRS enhanced host body weight very significantly than single mutant 923/coa(P<0.01);(2)Deletion mutants 923/coa,923/saeRS and 923/coa/saeRS had very significant lower cell counts in infection site than wild-type 923(P<0.01);(3)Infected mice serum IL-17a concentration increased after inoculation,and achieved to peak concentration in 24 hours,then gradually declined,and 8 h IL-17a concentration in wild type 923 infected mice serum was significantlg lower than 923/saeRS infected group.(P<0.05);(4)Mutant strains 923/saeRS and 923/coa/saeRS caused significantly smaller lesion size than wild type 923 and mutant strain 923/coa(P<0.05);(5)The correlation coefficient between 120 h skin lesion size and 24h serum IL-17a concentrations was significantly higher.It is concluded,therefore,that the deletion of coa and saeRS gene reduced the toxicity of wild-type strain 923,increased host body weight,decreased the bacterial counts and lesion size in infected areas,reduced the concentration of serum IL-17a.
