摘 要:肿瘤标志物的准确测量对肿瘤的早期筛查和预后评判具有重要意义。然而,由于复杂基质中肿瘤标志物含量极低,且易受高丰度蛋白质的信号干扰,因此对其直接质谱分析存在极大困难。本文将一种固载了特异性抗体的磁性纳米材料应用于肿瘤标志物热休克蛋白90α(Heat shock protein 90alpha,HSP90α)的分离富集,然后采用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF)分析HSP90α富集后的酶切肽段。并用实验验证该方法具有高效率、高特异性和高灵敏度的特点,该分析策略在临床肿瘤标志物的精准检测方面具有广阔的应用前景。The accurate measurement of tumor markers had great significance for early screening and prognostic evaluation of tumors. However,the direct mass spectrometric analysis was complicated due to the extremely low level of tumor markers in complex matrices and the susceptibility to signal interference from high-abundance proteins. In this paper,a magnetic nanomaterial loaded with specific antibodies was applied to the separation and enrichment of tumor marker heat shock protein 90α( HSP90α),followed by the analysis of the enrichment of digested peptides by matrix-assisted laser desorption ionization time-of-flight mass spectrometry( MALDI-TOF). The experimental results show that the developed method has higher efficiency,higher specificity,and higher sensitivity,and the analysis strategy has broad application prospects in the accurate detection of clinical tumor markers.
