摘  要:酵母菌的种群数量增长实验是高中生物教学中的一个综合性探究实验,是课程标准要求中的重要部分。本文对课程要求实验方法进行丰富和改进,以自制果酒分离获得的酵母菌株为实验材料,进行18S rDNA分子鉴定。在预实验的基础上,对酵母菌进行液体培养,利用分光光度计测定种群数量动态变化并绘制其生长曲线,分析酿酒酵母的生长特性及影响因素。The population growth experiment of Saccharomyces cerevisiae is a comprehensive exploration experiment in biology teaching of senior school.It is an important part of the curriculum standard.In this paper,the experimental methods required by the course were enriched and improved.Yeast strains isolated from fruit wine were used as experimental materials for 18S rDNA molecular identification.On the basis of the pre-experiment,the yeast was cultured in liquid medium,the population dynamic change was measured by spectrophotometer and its growth curve was drawn,and the growth characteristics and influencing factors of Saccharomyces cerevisiae were analyzed.
