摘 要:目的:观察丁氏妇科补肾养血保胎方(下简称保胎方)对肾虚型先兆流产大鼠免疫因子及凝血功能的影响。方法:将肾虚型先兆流产模型大鼠随机分为模型组、保胎灵组和保胎方低、中、高剂量组,药物干预10 d,另设正常组。检测血清IFN-γ、IL-2、IL-4、IL-10、PT、Fbg含量,观察各组体质量变化、子宫卵巢形态质量、胚胎数。结果:保胎灵组和保胎方中、高剂量组体质量变化及子宫卵巢质量大于模型组(P<0.05);保胎方中、高剂量组体质量变化大于保胎方低剂量组(P<0.01);保胎灵组和保胎方低、中、高剂量组正常胚胎数均高于模型组;保胎灵组和保胎方中、高剂量组活胎率均高于模型组。保胎灵组和保胎方低、中、高剂量组IFN-γ、IL-2表达均低于模型组(P<0.05);保胎灵组和保胎方低、中、高剂量组IL-10表达均高于模型组(P<0.05);保胎方中剂量组IL-4表达高于模型组(P<0.05);保胎方中剂量组IL-10表达高于保胎方低剂量组(P<0.01)。保胎灵组和保胎方低、中、高剂量组IL-4/IFN-γ、IL-10/IFN-γ、IL-10/IL-2均高于模型组(P<0.05);保胎方低、中剂量组IL-4/IL-2均高于模型组(P<0.05);保胎方中剂量组IL-10/IFN-γ高于保胎方低、高剂量组(P<0.05)。保胎方低、中、高剂量组PT长于模型组,保胎方低、中剂量组和保胎灵组Fbg含量低于模型组(P<0.05);保胎方中剂量组PT长于保胎方低、高剂量组(P<0.01),保胎方低、中剂量组Fbg含量低于保胎方高剂量组(P<0.05)。结论:保胎方可降低肾虚型先兆流产大鼠血清Th1细胞因子、提高Th2细胞因子表达,调整Th2/Th1比值偏向Th2;可延长PT、降低Fbg含量,降低血黏度;可提高其体质量、子宫卵巢质量,减轻胚胎与子宫损伤,提高保胎率;中剂量保胎方对多项指标改善有较理想的趋势。Objective: To observe the effects of Ding’s gynecological Bushen Yangxue Baotai prescription(hereinafter referred to as Baotai prescription) on immune factors and coagulation function in rats with threatened abortion due to kidney deficiency. Methods: The kidney-deficiency threatened abortion model rats were randomly divided into model group, Baotailing(保胎灵) group and Baotai prescription low, medium and high dose groups.Drug intervention for 10 days, and another normal group. Serum IFN-γ, IL-2, IL-4, IL-10, PT, Fbg levels were detected, and the changes in body mass, uterine and ovary morphology, and number of embryos were observed. Results: The changes of body mass and the uterine and ovarian mass of the Baotailing group and the Baotai prescription medium and high dose groups were greater than those of the model group(P<0.05). The body mass changes of the Baotai prescription medium and high dose groups were greater than those of the Baotai prescription low dose group(P <0.01). The number of normal embryos in the Baotailing group and the Baotai prescription low, medium and high dose groups were higher than that of the model group. The live birth rate of the Baotailing group and Baotai prescription medium and high dose groups were higher than that of the model group. The expression of IFN-γ and IL-2 in Baotailing group and Baotai prescription low, medium and high dose groups were lower than model group(P <0.05). The expression of IL-10 in Baotailing group and Baotai prescription low, medium and high dose groups was higher than that of the model group(P <0.05), and the expression of IL-4 in Baotai prescription medium dose group was higher than that of the model group(P<0.05),and the expression of IL-10 in Baotai prescription medium dose group was lower than that of Baotai prescription low dose group(P<0.01).
