摘 要:目的:探究复方七芍降压片降压作用机制。方法:SHR随机分为高血压对照组、抑制剂组、复方七芍组、抑制剂复方七芍组、厄贝沙坦组、抑制剂厄贝沙坦组,另取Wistar-Kyoto正常大鼠为正常对照组,采用免疫组化检测血管壁P2Y6、P65蛋白表达。大鼠血管平滑肌细胞随机分为空白对照组、抑制剂组、炎症组、抑制剂炎症组、含药血清组、抑制剂含药血清组,qPCR检测P65、P2Y6、IκBα基因表达,Western blotting检测P65、P2Y6、IκBα、p-IκBα蛋白表达。结果:动物实验结果显示,MRS2578对P2Y6表达无影响,使P65表达降低;复方七芍降压片使P2Y6和P65表达降低,与MRS2578联用可使P65表达更低;厄贝沙坦使P2Y6和P65表达均降低,与MRS2578联用可使P65表达更低。细胞实验结果显示,与空白对照组比较,TNF-α使P2Y6、P65、p-IκBα表达升高(P<0.05)。与TNF-α组比较,MRS2578对P2Y6无影响(P>0.05),使P65、p-IκBα表达降低(P<0.05);复方七芍降压片含药血清使P2Y6、P65、p-IκBα表达降低(P<0.05),与MRS2578联用可使P65、p-IκBα表达更低(P<0.05)。所有分组对IκBα表达无影响(P>0.05)。结论:复方七芍降压片可能通过抑制P2Y6的表达发挥降压功能,而机制可能是P2Y6表达降低抑制了NFκB信号通路的激活。Objective: To explore the antihypertensive mechanism of Fufang Qishao Jiangya tablets. Methods:SHR was randomly divided into blank group, inhibitor group, Fufang Qishao group, inhibitor +Fufang Qishao group, irbesartan group, and inhibitor + irbesartan group. Another Wistar-Kyoto normal rats was taken as the normal control group. Immunohistochemistry was used to detect the expression of P2Y6 and P65 protein in blood vessel wall. Rat vascular smooth muscle cells(VSMCs) were randomly divided into blank control group,inhibitor group, inflammation group, inhibitor+inflammation group, drug-containing serum group, and inhibitor +drug-containing serum group. The expression of P65, P2Y6, IκBα gene was detected by qPCR, and the protein expression of P65, P2Y6, IκBα, p-IκBα was detected by Western blotting. Results: The results of animal experiments show that MRS2578 has no effect on the expression of P2Y6 and can reduce the expression of P65. The Fufang Qishao Jiangya tablets reduces the expression of P2Y6 and P65, and the expression of P65 was lower when combination with MRS2578. Irbesartan reduces the expression of P2Y6 and P65, and the expression of P65 was lower when combination with MRS2578. The cell experiment results showed that compared with the blank control group, TNF-α increased the expression of P2Y6, P65, and p-IκBα(P<0.05). Compared with the TNF-α group, MRS2578 had no effect on P2Y6(P>0.05), and decreased the expression of P65 and p-IκBα(P<0.05). The serum containing Fufang Qishao Jiangya tablets reduced the expression of P2Y6, P65, and p-IκBα(P<0.05), and the expression of P65 and p-IκBαwas lower when combination with MRS2578(P<0.05). All groups had no effect on the expression of IκBα(P>0.05). Conclusion: Fufang Qishao Jiangya tablets may exert its antihypertensive function by inhibiting the expression of P2Y6, and the mechanism may be that the decrease of P2Y6 expression inhibits the activation of NFκB signaling pathway.
