摘　　要：目的探讨LncRNA WTAPP1在血管内皮祖细胞(EPCs)成血管中的作用及机制研究。方法构建LncRNA WTAPP1慢病毒载体,转染EPCs,将细胞分为3组:对照组、空载体组和转染组。RT-PCR法验证转染效率,四甲基偶氮唑蓝(MTT)法检测细胞存活率,Transwell实验检测细胞侵袭能力,体外成管实验检测细胞的成管能力。结果RT-PCR检测结果显示,转染后LncRNA WTAPP1表达水平显著上调,差异有统计学意义(P<0.05);MTT实验结果显示,转染组细胞生长速度明显高于对照组和空载体组,差异有统计学意义(P<0.05);Transwell结果显示,转染组细胞的侵袭能力明显高于对照组和空载体组,差异有统计学意义(P<0.05);成管实验结果显示,转染组细胞的成管能力明显高于对照组和空载体组,差异有统计学意义(P<0.05)。结论过表达LncRNA WTAPP1可增强EPCs增殖活性、侵袭和成血管能力。Objective To explore the role and mechanism of LncRNA WTAPP1 in vascular endothelial progenitor cells(EPCs).Methods The LncRNA WTAPP1 lentiviral vector was constructed and transfected into EPCs.The cells were divided into 3 groups:control group,empty vector group and transfection group.The transfection efficiency was verified by the RT-PCR method,the cell viability was detected by the tetramethylazazole blue(MTT)method,the cell invasion ability was detected by the Transwell experiment,and the tube formation ability of the cells was detected by the in vitro tube formation experiment.Results The results of RT-PCR showed that the expression of LncRNA WTAPP1 was significantly up-regulated after transfection(P<0.05).MTT results showed that the growth rate of the transfected group was significantly higher than that of the control group and the vector group(P<0.05).The results of Transwell showed that the invasiveness of the cells in the transfection group was significantly higher than that in the control group and the empty body group(P<0.05);the results of the tube forming experiment showed that the ability of the cells in the transfection group was significantly higher than that in the control group and the empty body group(P<0.05).Conclusion Overexpression of LncRNA WTAPP1 enhanced the proliferation,invasion and angiogenesis of EPCs.
