摘 要:以3个甘薯品种(北京553,烟薯25,卢选1号)为试验材料,研究外植体消毒和不同激素配比对茎尖诱导的影响。以甘薯茎尖分生组织为培养对象,筛选出适合甘薯不同品种的消毒方法和适合外植体生长的激素配方。结果表明,烟薯25,卢选1号,北京553分别用ρ(NaClO)=20.0 g·L^-1消毒9,13,11 min污染率最低,成苗率最高。适合烟薯25,卢选1号和北京553的最佳培养基分别是MS+1.5 mg/L 6-BA+0.1 mg/L IBA,MS+1.0 mg/L 6-BA+0.1 mg/L IBA和MS+1.0 mg/L 6-BA+0.5 mg/L IBA。Three sweet potato varieties,including Beijing-553,Yanshu-25 and Luxuan-1,were used as materials to reveal the effects of explant sterilization and different hormone combinations on the induction of shoot tip.The meristem of the stem tip of sweet potato was used as the object of culture to select suitable disinfection method for various varieties of sweet potato and hormone combination for explant growth.The results showed that the contamination rate was the lowest and the seedling rate was the highest when Yanshu-25,Luxuan-1 and Beijing-553 were sterilized byρ(NaClO)=20.0 g·L^-1 for 9 min,13 min and 11 min,respectively.The best culture medium for Yanshu-25,Luxuan-1 and Beijing-553 were MS+1.5 mg/L 6-BA+0.1 mg/L IBA,MS+1.0 mg/L 6-BA+0.1 mg/L IBA,MS+1.0 mg/L 6-BA+0.5 mg/L IBA,respectively.
