摘 要:试验旨在探究高繁殖力候选基因在和田羊群体的遗传特征,为和田羊多羔类群的选育奠定基础。以不同产羔数的和田羊为研究对象,借助特异性内切酶,对TGFβ/BMP信号通路中GDF9(G1位点)、BMPR1B(FecB位点)与BMP15基因(FecXG位点)进行酶切位点分析。经检测,在和田羊中首次发现GDF9、BMPR1B基因具有多态性,GDF9存在2种基因型(GG、GA),BMPR1B存在3种基因型(++、+B、BB),而BMP15基因无多态性;GDF9、BMPR1B基因的PIC分别为低度多态、中度多态,均处于Hardy-Weinberg平衡状态(P>0.05),可世代稳定遗传;GDF9、BMPR1B基因在和田羊母羊群体中存在4种组合基因型,在公羊群体中存在2种组合基因型,公羊、母羊群体的遗传结构特征可能存在差异。以上结果揭示,BMP15基因不是影响和田羊繁殖性状的候选基因;GDF9、BMPR1B基因有待与和田羊繁殖性状进行关联分析以进一步确认。The experiment aimed to explore the genetic characteristics of candidate genes with high fertility in Hotan sheep population,lay the foundation for the breeding of Hotan Sheep with multiple lambs.In this paper,the RFLP analysis of GDF9 gene(G1 site),BMPR1B gene(FecB site)and BMP15gene(FecXG site)of TGFβ/BMP signaling pathway was carried out by using specific endonuclease.It was first found that GDF9 gene had two genotypes(GG,GA),BMPR1B gene had three genotypes(++,+B and BB),and there was no polymorphism in BMP15 gene.The PIC of GDF9 and BMPR1B gene were in low and medium degree polymorphism,respectively,and both of them were in Hardy-Weinberg equilibrium(P>0.05),which meant the two genes could be inherited stably for generations.There were 4 combined genotypes of GDF9 and BMPR1B gene in ewe group of Hotan sheep and 2 combined genotypes in ram group,indicating that the genetic structure characteristics of ram and ewe group may be different.As a conclusion,BMP15 gene was not the candidate gene affecting the reproductive traits of Hotan sheep,while GDF9 and BMPR1B gene need to be further confirmed by correlation analysis with reproductive traits.
