摘  要:越来越多的证据表明,除经典的遗传控制外,表观遗传调控也参与到众多的细胞发育过程。雌性果蝇幼虫性腺的发育涉及细胞增殖及分化,是良好细胞生物学在体研究模型。文章通过果蝇遗传学实验方法,研究发现ATP依赖的染色质重塑因子Brahma(Brm)参与调控雌性果蝇幼虫原始生殖细胞(PGC)有序分化的证据,brm基因突变的三龄幼虫雌性果蝇,PGC的分化被提前开启;此外,特异性的将PGC中的brm表达量下调,也发现PGC的提前分化。这说明,表观遗传调控因子Brm参与调控雌性果蝇幼虫性腺发育,并以细胞自治性的方式调控原始生殖细胞的有序分化。Besides the genetic regulation,epigenetic mechanisms also regulate cell development.Drosophila larval ovary morphogenesis mainly involves primordial germ cell(PGC)proliferation and differentiation,which has become an in vivo working platform for studying cell biology.Using genetic experiments in Drosophila,we found that the Brahma(Brm),the ATPase subunit of the Drosophila SWI/SNF chromatin-remodeling complexes,is required for PGC program differentiation.The removal or knockdown of brm function in germline promoted PGC differentiation at late 3 rd instar larvae(LL3).These results show that Brm is essential for Drosophila larval PGC program differentiation cell-autonomously.
