摘  要:目的探讨小鼠颅脑爆震伤的早期评估指标。方法将20只小鼠平均分为正常对照组和创伤性脑损伤(bTBI)模型组,每组各10只。建立爆震伤动物模型后15 d内,观察小鼠脑部经冲击波暴露后意识丧失持续时间、创伤后记忆缺失持续时间等表现;采用HE染色、蛋白质印迹法等方法对小鼠脑组织病理改变进行分析,观察两组小鼠脑组织中的S-100β、髓鞘碱性蛋白(MBP)、神经元特异性烯醇化酶(NSE)指标变化及mRNA的表达情况。结果建模后,bTBI模型组小鼠无死亡。建模3 h内,小鼠均清醒,行动较迟缓,4 h小鼠进食、水,活动速度有所增加,但与正常对照组相比较,活动较迟缓。建模后7、14 d,bTBI模型组小鼠进入平台的次数均明显少于正常对照组,两组间比较,差异均有统计学意义(P<0.05)。建模后7、14 d,正常对照组小鼠找到平台位置的时间均明显短于bTBI模型组,两组间比较,差异有统计学意义(P<0.05)。小鼠的活动轨迹显示,正常对照组小鼠围绕平台的活动轨迹较规律,bTBI模型组的活动轨迹相对较分散。解剖后,bTBI模型组小鼠的脑组织边缘较钝,组织边缘棱角不清晰,有明显的水肿表现;正常对照组脑组织体积明显缩小,组织边缘清晰,可见明显的血管及组织结构。正常对照组小鼠海马区神经元细胞排列规则,无损伤;实验小鼠的脑组织中海马区神经元发生了变性,同时锥体细胞发生肿胀且不规则排列。bTBI模型组小鼠脑组织氧化应激指标s100β、NSE和MBP的表达量均显著高于正常对照组,差异均有统计学意义(P<0.05)。结论bTBI可降低小鼠的认知能力、损伤小鼠的海马区神经元等。通过建立bTBI早期筛选评估模型可知,检测大脑标志物s100β、NSE和MBP的表达变化,有助于对早期bTBI进行快速筛选识别。Objective To investigate the early evaluation index of brain blast injury in mice.Methods A total of 20 cases of mice were divided into the normal control group and the blast-induced traumatic brain injury(bTBI)establishment group,with 10 in each group.The duration of consciousness loss and memory loss after shock wave exposure in the brain of mice within 15 days after detonation injury were observed.HE staining and Western-blot were used to analyze the pathological changes of mouse brain tissue,and the changes of S-100β,myelin basic protein(MBP),neuron-specific enolase(NSE),and mRNA expression in the brain tissue of the two groups of mice were observed.Results After establishment,no death occurred in the bTBI establishment group.Within 3 hours of establishment,the mice were awake and moved slowly,while 4 hours of feeding and water increased the activity speed of the mice,but the activity was slower compared with the normal control group.After 7 and 14 days of establishment,the times of mice entering the platform in the bTBI establishment group were significantly lower than those in the normal control group,and the differences between the two groups were statistically significant(P<0.05).After 7 and 14 days of establishment,the times for mice in the normal control group to find the platform location were significantly shorter than those in the bTBI establishment group(P<0.05).The activity trajectories of mice showed that the normal control group had regular trajectories around the platform,while the bTBI establishment group had relatively scattered trajectories.After anatomy,the brain tissue edges of the bTBI establishment group mice were blunt,with unclear edges and corners,showing obvious edema.In the normal control group,the brain tissue volume was significantly reduced,the tissue margins were clear,and obvious blood vessels and tissue structures could be seen.In normal control group,the neurons in hippocampus were arranged in a regular way without injury.The hippocampal neurons denaturated and pyramidal cell
