摘 要:目的:采用高效液相色谱法对不同产地的穿山龙中薯蓣皂苷元含量进行测定。方法:选取高效液相色谱法作为含量测定方法,色谱柱为Diamonsil C18(4.6 mm×250.0 mm,5μm),流动相选择乙腈-水(5545),流速控制在1.0 mL·min^-1,检测波长203 nm。对不同产地的穿山龙进行薯蓣皂苷元含量的测定,重复测定3次。结果:实验结束后,分别获得3.01%、3.18%、3.42%、2.86%、2.20%、1.41%的结果,平均含量2.68%。结论:采用乙腈-水(5545)作为流动相,获取了较为良好的薯蓣皂苷元色谱峰峰形,保留时间较长。采用高效液相色谱法对不同产地的穿山龙中薯蓣皂苷元含量进行测定,供试品溶液基本保持稳定,具备良好的精密度和线性关系,且重现性好,适用于该生药及提取物的质量控制。Objective:To determine the content of dioscin in Chuanshanlong(Rhizoma Dioscoreae Nipponicae)by high-performance liquid chromatography(HPLC).Methods:HPLC was used as the content determination method.The chromatographic column was diamonsil CIG(4.6 mm×250 mm,5μm);the mobile phase was acetonitrile water(55:45);the flow rate was controlled at 1.0 mL·min-1,and the detection wavelength was 203 nm.Dioscin content of Chuanshanlong from different habitats was determined respectively,and the determination was repeated for 3 times.Results:After the experiment,3.01%,3.18%,3.42%,2.86%,2.20%and 1.41%were obtained respectively,with an average content of 2.68%.Conclusion:Using acetonitrile water(55:45)as mobile phase,the better chromatographic peak shape of dioscin can be obtained,and the retention time is longer.If the content of dioscin in Chuanshanlong from different habitats is determined by HPLC,the test solution will be basically stable,with good precision and linear relationship and good reproducibility,which is suitable for the quality control of raw Chuanshanlong and its extracts.
