摘 要:目的探讨保肾通络方对KK-Ay小鼠肾脏足细胞p38通路及凋亡的影响,进而阐明保肾通络方防治DN的分子机制。方法30只KK-Ay小鼠随机分为模型组、保肾通络方组及缬沙坦组,另选取10只C57BL/6J小鼠作为正常对照组。保肾通络方组和缬沙坦组分别给予保肾通络方和缬沙坦灌胃,正常对照组和模型组予等剂量蒸馏水灌胃,实验周期为12周。结果与正常对照组相比,模型组足细胞p38通路显著激活,足细胞凋亡数目明显增加(P<0.05);与模型组相比,保肾通络方组及缬沙坦组小鼠足细胞p38通路激活被抑制,足细胞凋亡数目明显减少(P<0.05)。结论保肾通络方可以抑制DN小鼠足细胞p38通路激活,减轻足细胞凋亡,可能是其降低DN蛋白尿,延缓疾病进展的分子机制。Objective To observe the effect of Baoshen Tongluo Formula(BSF)on podocye apoptosis and p38 pathway in DN mice.Methods The model KK-Ay mice were separated into 3 groups randomly:DN group,Valsartan group and BSF group.Ten C57 mice performed for NC group.Mice in BSF group were given BSF.Mice in Valsartan group were given Valsartan.Mice in DN and NC group were given normal saline.All mice were successively given the administration for 12 weeks.Results As compared with NC group,p38 pathway was significantly activated and podocyte apoptosis was significantly increased in DN group(P<0.05).Compared with DN group,P-p38 expression and podocyte apoptosis was significantly decreased in Valsartan group and BSF group(P>0.05).Conclusion BSF can alleviate proteinuria and ameliorate renal function of DN,which partially through inhibiting p38 pathway and decreasing podocyte apoptosis in DN.
