摘 要:NAC转录因子是植物特有的、最大转录因子家族之一,参与植物生长发育和逆境胁迫应答。本研究克隆了中间锦鸡儿CiNAC071基因,该基因由2个内含子和3个外显子构成,开放阅读框为1 017 bp,编码338个氨基酸。在CiNAC071过表达拟南芥荧光定量分析发现:CiNAC071通过下调SOS1、NHX和RD22基因的表达,增强过表达植物的盐敏感性且降低鲜重。同时,经2次PCR扩增共获得2 108 bp的启动子序列。经在线分析,该启动子具有脱落酸(abscisic acid, ABA)、茉莉酸甲酯(methyl jasmonate, MeJA)、赤霉素(gibberellin, GA)和低温诱导等顺式作用元件。将CiNAC071启动子构建表达载体并转染野生型拟南芥,启动子驱动GUS报告基因主要集中在拟南芥子叶叶脉、新生叶片、下胚轴和花萼顶端的叶脉中表达;且CiNAC071启动子为诱导型启动子, ABA处理抑制GUS表达, GA处理促进GUS表达。NAC transcription factor family is the plant-specific and one of the largest transcription factor families, and it is involved in plant growth, development and stress responses. In this study, Caragana intermedia CiNAC071 genes was cloned, and it contains two introns and three exons. Its open reading frame was 1 017 bp, encoding 338 amino acids. Quantitative analysis of CiNAC071-overexpressing Arabidopsis thaliana lines showed that CiNAC071 enhanced plants salt sensitivity and reduced fresh weight by down-regulating the expression of SOS1, NHX and RD22 genes. At the same time, its promoter sequence, which was 2 108 bp, was obtained by two PCR amplifications. After online analysis, we knew that the promoter had many cis-elements, such as ABA, MeJA, GA, low-temperature responsive cis-elements and the others. The CiNAC071 promoter was used to construct an expression vector and transfected into wild-type Arabidopsis, and the GUS reporter activity driven by the promoter was mainly concentrated in Arabidopsis cotyledon veins, young leaves, hypocotyl and leaf veins at the top of the calyces. And the promoter was inducible, which was inhibited by ABA but induced by GA treatment.
