摘  要:本研究旨在鉴定牦牛疑似脑多头蚴病的病原,通过PCR分子生物学方法扩增了囊泡状样品的cytb基因和nad4基因序列,并进行分子鉴定与系统发育分析。结果:经分子鉴定,病原确诊为脑多头蚴;克隆的cytb和nad4基因序列分别与GenBank上的多头带绦虫参考序列的同源性达到98.7%和99.2%以上;利用GenBank中其他带属绦虫的cytb和nad4基因序列构建系统发育树,均与已鉴定的多头带绦虫聚在同一支上,但并未形成微小分支,与其它绦虫所属分支相距较远。本研究初步分析了脑多头蚴青海分离株与其他种属之间的系统发育关系,丰富了脑多头蚴的群体遗传学研究资料,为牦牛脑多头蚴病的诊断研究等提供科学参考。In order to identify the pathogen of the suspected coenuriasis in the brain of the yak,the PCR method was carried out to amplify cytb and nad4 gene partial sequences from vesicle-like sample.Their homologies with genes from some other tapeworm species were analyzed and the phylogenetic trees were constructed,respectively.The results showed that the parasite was identified as Coenurus cerebralis actually.The cloned cytb and nad4 gene homogeneity was above 98.7%and 99.2%with T.multiceps reference sequences on the Genbank,respectively.Meanwhile,the cytb and nad4 gene sequences were used to construct phylogenetic trees with those of other tapeworm species from the GenBank database.It was found that the collected tapeworm was in the same branch with T.multiceps identified in the two phylogenetic trees,but it did not form tiny branches,which were separated from the branches of other tapeworm species,and the phylogenetic relationship between the identified T.multiceps and other tapeworm species was elucidated.This study enriched the population genetics research data of T.multiceps,and provided a scientific basis for the diagnosis of this diseases.
