摘  要:目的:研究蒙古黄芪总皂苷对铅染毒大鼠的学习记忆损伤的保护作用。方法:将45只4周龄清洁级SD大鼠用完全随机法等分为阴性组、模型组、蒙古黄芪总皂苷高(0.1500 g/Kg)、中(0.0750 g/Kg)、低(0.0375 g/Kg)剂量组;通过水迷宫实验评估大鼠的学习记忆能力;通过酶联免疫吸附实验检测血清丙二醛(MDA)、超氧化物歧化酶(SOD)和脑源性神经营养因子(BDNF)含量。结果:水迷宫测试显示,与模型组比较,蒙古黄芪总皂苷高、中、低剂量组第一次到达平台所用时间短,在平台及其所在象限停留时间比率高,进入平台所在位置次数多,差别有统计学意义(P<0.05)。与模型组比较,蒙古黄芪总皂苷血清中MDA含量降低,SOD、BDNF含量增加,差异有统计学意义(P<0.05)。结论:蒙古黄芪总皂苷可能是通过降低血清MDA含量,增加SOD、BDNF含量发挥抗氧化作用和神经保护作用,提高铅暴露大鼠学习记忆能力。Objective:To study the protective effect of Mongolian Astragalus saponins on learning and memory impairment in rats exposed to lead.Methods:Fifty four-week-old clean SD rats were divided into negative group,model group,Mongolian Astragalus total saponin high(0.1500g/Kg),medium(0.0750g/Kg),low(0.0375g/Kg)dose group;the learning and memory ability of rats was evaluated by water maze test;serum malondialdehyde(MDA)and superoxide dismutase were detected by enzyme-linked immunosorbent assay(SOD)and neurotrophic factor(BDNF)content.Results:Water maze results:Compared with the model group,the Mongolian Astragalus total saponin high-medium and low-dose group took a short time to reach the platform for the first time,had a high ratio of residence time on the platform and its quadrant,and entered the platform more times and the difference was statistically significant(P<0.05).Compared with the model group,the serum MDA content of Mongolian Astragalus saponins decreased,and the content of SOD and BDNF increased.The difference was statistically significant(P<0.05).Conclusion:The total saponins of Astragalus mongholicus may play an antioxidant and neuroprotective role by reducing the content of serum MDA,increasing the content of SOD and BDNF,and improving the learning and memory ability of rats exposed to lead.
