摘 要:目的探讨妊娠期糖尿病(GDM) SD大鼠对子代大鼠心脏缺血再灌注(I/R)损伤的影响及作用机制。方法选择妊娠中期SD雌性大鼠25只,按照随机数字表法分为研究组(13只,腹腔注射35 mg/kg链脲佐菌素建立GDM模型)和对照组(12只,腹腔注射等量柠檬酸盐缓冲液)。对比两组子代大鼠刚出生时的体重,饲喂24周后的体重、全心质量和体重与全心质量比值;对两组子代大鼠构建心脏离体I/R模型,生理计录仪测定I/R前和I/R后大鼠心脏功能指标;western blot检测两组子代大鼠I/R前和I/R后心肌相关蛋白激酶B(PKB)、腺苷酸激活蛋白激酶(AMPK)、乙酰辅酶A羧化酶(ACC)的相对表达量。结果 (1)两组子代大鼠刚出生时的体重比较,差异无统计学意义(P>0.05);生长至24周时,子代研究组体重(286.91±18.17 vs.257.91±19.02) g、全心质量(850.12±130.41 vs.718.94±124.57) mg、全心质量/体重(2.95±0.12 vs.2.78±0.10) mg/g,明显高于子代对照组(P<0.05)。(2)I/R后,子代研究组心率与收缩压的乘积(RPP)水平(3 350.84±319.26 vs.6 710.12±424.91)次/min×mm Hg,明显低于对照组,差异具有统计学意义(P<0.05);(3)I/R后,两组ACC蛋白(研究组:1.32±0.24 vs.0.68±0.09;对照组:1.87±0.41 vs.0.69±0.12)和AMPK蛋白(研究组:2.28±0.51 vs.1.13±0.33;对照组:3.39±0.58 vs.1.12±0.22)相对表达量均较I/R前明显升高,且子代研究组明显低于子代对照组,差异具有统计学意义(P<0.05)。结论 GDM大鼠对子代的生长发育过程会产生影响,导致体重增加和心脏功能异常及对子代大鼠心脏I/R损伤产生不耐受,从而引起心脏功能异常。Objective To investigate the effects and mechanism of gestational diabetes (GDM) SD rats on the offspring’s heart ischemia-reperfusion (I/R) injury.Methods Twenty-five SD female rats in mid-pregnancy were selected and divided into a research group (13 rats with intraperitoneal injection of 35 mg/kg streptozotocin to establish GDM model) and a control group (12 rats with intraperitoneal injection of equal amounts of lemon Salt buffer) according to the random number table method.Body mass at birth was compared,and body mass,total heart mass and total heart mass ratio after 24 w feeding were compared between the two groups.The isolated I/R model of heart was constructed in two groups of offspring rats,and physiograph to measure the rat heart function indexes before and after I/R;Western blot was used to detect the relative expression of myocardial-associated protein kinase B (PKB),adenylate-activated protein kinase (AMPK),and acetyl-CoA carboxylase(ACC) before and after I/R.Results (1)When growing to 24 weeks,body mass (286.91±18.17 vs.257.91±19.02) g,whole heart mass(850.12±130.41 vs.718.94±124.57) mg and whole heart mass/body mass (2.95±0.12 vs.2.78±0.10) mg/g of the offspring in study group were significantly higher than those of the control group (P<0.05).(2)After I/R,the RPP level (3 350.84±319.26 vs.6710.12±424.91)times/min×mm Hg of the study group was significantly lower than that of the control group (P<0.05).(3)There was no significant difference in the relative expression of PKB protein between the two groups before I/R and after I/R (P>0.05).After I/R,the relative expression levels of ACC(study group:1.32±0.24 vs.0.68±0.09;control group:1.87±0.41 vs.0.69±0.12) and AMPK protein(study group:2.28±0.51 vs.1.13±0.33;control group:3.39±0.58 vs.1.12±0.22) in both groups were significantly increased,and the subgroup in study group was significantly lower than that of the subgroup in control group (P<0.05).Conclusion GDM rats can affect the growth and development of their progeny,resulting in in
