摘 要:目的探讨Shh信号通路抑制剂环巴胺对人直肠癌细胞裸鼠皮下移植瘤形成的影响。方法取60只裸鼠,随机分为对照组、同时给药组、延迟给药组各20只,三组均于左后腿外侧皮下注射直肠癌细胞。注射癌细胞后,对照组立即给予生理盐水0.2 mL皮下注射,同时给药组立即给予20μmol/L环巴胺0.2 mL皮下注射;注射癌细胞5 d后,延迟给药组肉眼可见肿瘤长出,给予环巴胺20μmol/L环巴胺0.2 mL皮下注射。三组于给药10、20 d分别处死10只大鼠,比较移植瘤体积抑制率、移植瘤质量抑制率和移植瘤细胞密度。结果与延迟给药组相比,同时给药组10 d、20 d后的移植瘤体积抑制率、质量抑制率均升高,移植瘤细胞密度均降低(P均<0.05)。与给药10 d相比,同时给药组、延迟给药组20 d后的移植瘤体积抑制率、质量抑制率均升高,移植瘤细胞密度均降低(P均<0.01)。结论环巴胺不同时间应用均可抑制裸鼠直肠癌移植瘤生长,可用于直肠癌的靶向治疗。Objective To investigate the effect of cyclopamine inhibitor cyclopamine on subcutaneous transplantation of human rectal cancer cells in nude mice.Methods Sixty nude mice were randomly divided into the control group,simultaneous administration group and delayed drug-administration group.The rectal cancer cells were subcutaneously injected into the lateral sides of the left hind legs of nude mice in the three groups.After the injection of cancer cells,the mice in the control group were given subcutaneous injection of 0.2 mL normal saline immediately,and the administration group was given subcutaneous injection of cyclopamine 25 mg/(kg·d)immediately.At 5 days after the injection of cancer cells,the tumor was visually observed in the delayed drug-administration group,and the mice were given subcutaneous injection of 20μmol/L cyclopamine 0.2 mL.In the three groups,10 rats were sacrificed at 10 and 20 days after administration,and the volume inhibition rate of the transplanted tumor,the mass inhibition rate of the transplanted tumor and the density of the transplanted tumor cells were compared.Results Compared with the delayed drug-administration group,the volume inhibition rate and mass inhibition rate of the transplanted tumor increased at 10 and 20 days in the simultaneous administration group,and the cell density of the transplanted tumor decreased(all P<0.05).The volume inhibition rate and mass inhibition rate of transplanted tumors increased,and the cell density of transplanted tumors decreased in the simultaneous administration group and delayed drug-administration group at 20 days after administration as compared with those at 10 days after administration(all P<0.01).Conclusion Cyclopamine can inhibit the growth of transplanted tumors of rectal cancer in nude mice;early intervention or extension of treatment duration can have a better effect on tumor inhibition.
