摘  要:目的:为了快速检测早餐汤料中的罂粟壳生物碱残留,研究并建立一种利用表面增强拉曼光谱技术(Surface-enhanced Raman spectroscopy,SERS)检测罂粟壳中吗啡、可待因、罂粟碱、那可丁、蒂巴因5种生物碱的定性与定量方法。方法:采用MPFC-QuEChERS超滤净化柱对样品进行前处理,根据经典柠檬酸钠还原HAuCl4方法制备金溶胶,并对仪器参数进行优化,结合SERS技术采集5种罂粟壳生物碱的拉曼光谱特征峰并作为快速鉴定的定性依据,分别选取罂粟碱、那可丁、可待因、蒂巴因、吗啡在724、1274、600、1593、627 cm—1处谱峰进行定量分析。结果:加入制备的金溶胶后,罂粟碱、那可丁、可待因、蒂巴因、吗啡均出现明显的拉曼特征峰,其特征峰强度与质量浓度在一定范围内有良好的线性关系,其中检出限为0.0005μg/g,方法平均添加回收率为90.5%~98.4%,相对标准偏差(RSD)为5.2%~8.3%。结论:使用SERS技术迅速简便、绿色环保、确证性高,可实现对羊肉汤中罂粟壳残留进行定性与定量分析,在汤料类食品安全质量监控方面具有良好的应用潜力。Objective:In order to quickly detect the alkaloid residues of poppy shells in breakfast soup,a research and establishment of a surface-enhanced Raman spectroscopy technique for morphine,codeine,papaverine,nacodine and thebaine in poppy shell qualitative and quantitative methods were carried out.Methods:The MPFC-QuEChERS ultrafiltration purification column was used to perform solid-phase extraction pretreatment on the sample.Gold nano-reinforced substrate was prepared according to the classic sodium citrate reduction HAuCl4 method.The instrument parameters were optimized and five kinds of poppy shell alkaloids were collected in combination with SERS technology.The Raman spectral characteristics of the sample were used as a qualitative basis for rapid identification.Papaverine,noscapine,codeine,thebaine and morphine were selected for quantitative analysis at 724,1274,600,1593,627 cm—1 spectral peaks.Results:After adding the prepared gold colloid,papaverine,noscapine,codeine,thebaine and morphine all have obvious Raman characteristic peaks,and the characteristic peak intensity and mass concentration have a good linearity in a certain range.The minimum detection limit is 0.0005μg/g.Relationship,the average recovery of the method is between 90.5%~98.4%,and the relative standard deviation is 5.2%~8.3%.Conclusion:Studies have shown that the use of SERS technology is quick and easy,environmentally friendly,and highly conclusive.It can realize qualitative and quantitative analysis of poppy shell residues in mutton soup,and has good application potential in the safety and quality monitoring of soup foods.
