摘 要:目的研究西红花苷对子宫腺肌病异位子宫内膜间质细胞生长周期及凋亡的影响,从细胞水平初步探讨西红花苷在子宫腺肌病中的治疗作用。方法2018年1~6月收集江西省妇幼保健院术后病理确诊为子宫腺肌病标本,不同浓度西红花苷(0、200、300、400、500μg/mL)作用于异位子宫内膜间质细胞后,分别在培养0、24、48、72、96 h取出,CCK-8法检测细胞增殖;Annexin V-FITC/PI双染色流式细胞术检测细胞凋亡;PI单染检测细胞周期。结果CCK-8法检测西红花苷浓度分别为200、300、400、500μg/mL作用细胞72 h后,细胞抑制率分别为(7.60±0.03)%、(11.50±0.10)%、(18.26±0.14)%、(20.62±0.09)%,浓度为400μg/mL、500μg/mL组增殖抑制率明显高于浓度为200μg/mL组,结果显示不同浓度西红花苷对子宫腺肌病异位内膜细胞增殖有明显抑制作用,与药物浓度及时间呈正比,随着西红花苷浓度的升高和作用时间的延长,对细胞增殖的抑制率也越高;不同浓度西红花苷作用细胞48 h后,流式细胞术检测到子宫腺肌病异位子宫内膜间质细胞凋亡率随浓度的增加而增大,细胞周期G0/G1期比例增多。结论西红花苷在体外能显著抑制子宫腺肌病异位子宫内膜间质细胞增殖,诱导凋亡,使细胞周期阻滞在G0/G1期,这些结果在细胞水平上显示西红花苷可用于临床治疗子宫腺肌病。Objective To explore the impact of crocin on the growth cycle and apoptosis of ectopic endometrial stromal cells in adenomyosis,and to preliminarily investigate the therapeutic effect of crocin on adenomyosis from cells level.Methods From January to June 2018,specimens of adenomyosis confirmed by postoperative pathology in Jiangxi Maternal and Child Health Hospital were collected.After crocin(0,200,300,400,500μg/mL)of different concentrations performed on ectopic endometrial stromal cells,they were taken out after 0,24,48,72,96 hours of culture respectively,and cell proliferation was detected by CCK-8 method.Annexin V-FITC/PI double staining flow cytometry was used to detect cell apoptosis.PI single staining was used to detect cell cycle.Results The cell inhibition rates were(7.60±0.03)%,(11.50±0.10)%,(18.26±0.14)%,(20.62±0.09)%,respectively after different concentrations of crocin of 200,300,400,500μg/mL applied to the cells for 72 h detected by the CCK-8 method.The proliferation inhibition rate of 400μg/mL、500μg/mL were significantly higher than that of the 200μg/mL group.The results showed that crocin of different concentrations had obvious inhibition effect on the proliferation of ectopic endometrial cells of adenomyosis,which was proportional to the drug concentration and time.The higher of crocin concentration and the longer of acting time,the higher of the cell proliferation inhibition rate.48 hours after crocin at different concentrations acted on cells,flow cytometry detected that with the increase of concentration,the apoptosis rate of ectopic endometrial stromal cells in adenomyosis increased and the G0/G1 phase ratio of cell cycle increased.Conclusion Crocin can significantly inhibit proliferation of ectopic endometrial stromal cells of adenomyosis in vitro,induce apoptosis,lead cell cycle to G0/G1 phase,suggesting from cell levels that crocin can be used for clinical treatment of adenomyosis.
