摘  要:干细胞因来源广泛、可自我更新、拥有多向分化潜能等优点,目前已广泛用于组织修复实验和临床研究,特别是在骨缺损修复中展现出良好的应用前景。在骨组织工程中,干细胞作为缺损修复支架的组成要素,为缺损修复提供细胞来源和活性基础,因此探究骨缺损修复中干细胞成骨分化的主要调控因素尤为重要。本文将从支架的材料组成、材料表面微观结构、空间宏观结构、外源性药物、体外刺激等因素综述骨组织工程中干细胞成骨分化调控因素、作用机理与研究进展,为骨缺损修复的基础研究与临床应用提供新思路。Stem cells have been widely used in tissue repair experiments and clinical research specially showing good application prospects in bone defect repair,due to their advantages of wide source,self-renewal,and multi-directional differentiation potential.In bone tissue engineering,stem cells,as a component of defect repair scaffolds,provide cell source and active basis for defect repair.Therefore,it is particularly important to explore the main regulatory factors of stem cell osteogenic differentiation in bone defect repair.In this article,we will review the regulatory factors,mechanism and research progress of osteogenic differentiation of stem cells in bone tissue engineering from the factors such as the material composition of the scaffold,the microstructure of the surface of the material,the macroscopic structure of the space,the exogenous drugs,and in vitro stimulation.This provides a new idea for the basic research and clinical applications of bone defect repair.
