摘 要:利用条件性基因敲除方法和免疫荧光技术,检测和分析了小鼠从胚胎到成体耳蜗柯蒂氏器中Gata3的表达.结果表明,Gfi1-cre可以高效敲除特定时期毛细胞内Gata3基因的表达.同时发现成体Gata3-Null的毛细胞数量减少且排列发生紊乱.提示转录因子GATA3在耳蜗毛细胞发育和结构维持中起着重要作用.The conditional gene knockout(CKO)method and immunofluorescence technology are used to detect and analyze the expression of Gata3 in the Organ of Corti(OC)of the mouse cochlea from embryo to adult stage.The results show that Gfi1-cre can efficiently knock out Gata3 expression in hair cells at specific developmental stages.It is also found that the number and arrangement of hair cells in adult Gata3-null mice are disordered.It is suggested that the transcription factor GATA3 plays an important role in the development of cochlear hair cells and the maintenance of OC structure.
