猪血凝性脑脊髓炎病毒间接ELISA抗体检测试剂盒的研制与应用

摘　　要：以猪血凝性脑脊髓炎病毒(porcine hemagglutinating encephalomyelitis virus,PHEV)重组蛋白N(PHEV-N)为包被抗原,在建立间接ELISA抗体检测方法的基础上,进行了试剂盒组装及初步应用。结果显示,利用该试剂盒检测来自吉林、山东、天津、辽宁和黑龙江等地区部分猪场(前期曾有哺乳期发病仔猪表现神经症状和呕吐等)的血清样品,PHEV抗体阳性率分别为62.56%(127/203),85.96%(153/178),71.11%(96/135),53.74%(79/147),60.75%(113/186)。结果表明,该试剂盒兼具操作简便、快速、高通量等优势,不仅可用于PHEV感染猪群的快速诊断,而且可用于该病的血清学调查。Porcine hemagglutinating encephalomyelitis is an acute,highly contact infectious disease caused by porcine hemagglutinating encephalomyelitis virus(PHEV)infection in piglets.The clinical symptoms are similar to pig pseudorabies and infectious gastroenteritis.In order to make the clinical diagnosis of PHEV more accurate,this study use PHEV-N recombinant protein as the detection antigen to establishes an indirect ELISA antibody detection method,assemble of the kit and performs preliminary application.Using this standardized kit to detect clinical samples from some pig farms in Jilin,Shandong,Tianjin,Liaoning,and Heilongjiang provinces,the results showed that the positive rates of PHEV antibodies were 62.56%(127/203),85.96%(153/178),71.11%(96/135),53.74%(79/147),and 60.75%(113/186)respectively,suggesting that PHEV recessive infection is widespread in pig farms in above areas.The indirect ELISA antibody detection kit developed in this study has the advantages of simple operation,fast speed,and high throughput,and can be effectively applied to the serological diagnosis of PHEV infection in pigs.
