摘 要:对3株(GD1406、FS2015、FJFZ株)变异伪狂犬病病毒(PRV)的gB和gC全基因进行扩增、测序及生物信息学分析,应用微量交叉中和试验分析抗原差异性。基因序列比对结果显示,3株PRV变异株之间gB、gC基因核苷酸和氨基酸同源性分别为99.7%~100.0%和99.6%~100.0%,100.0%和99.0%~100.0%;gB和gC基因系统进化树遗传进化分析显示:系统进化树分为2大支,其中一个大支为国内经典毒株群、2011年后分离的变异毒株群2个小分支所占据的中国毒株群;另一个大支为欧洲分离株和美洲分离株组成的欧美毒株群。微量交叉中和试验结果显示,PRV-Bartha K61高免血清中和PRV-GD1406和PRV-Bartha K61的效价分别为22和53,PRV-GD1406高免血清中和PRV-GD1406和PRV-Bartha K61的效价分别为37和44;PRV-GD1406毒株异源血清中和效价/同源血清中和效价约为0.83,PRV-Bartha K61毒株异源血清中和效价/同源血清中和效价约为0.59;2个毒株(PRV-GD1406、PRV-Bartha K61)间交叉反应程度相关系数R=0.72。This study applied three strains(GD1406,FS2015,FJFZ strain)variant pseudorabies virus gB and gC.Whole-genome amplification,sequencing,and bioinformatics analysis were performed using micro-crossover neutralization assays for antigen differential analysis.The results of gene sequence alignment showed that the nucleotide and amino acid homology of the gB gene between the three strains of PRV were 99.7%-100.0%and 99.6%-100.0%,respectively;the gC gene between the three mutant pseudorabies nucleotide and amino acid homology were 100.0%and 99.0%-100.0%,respectively.gB gene and gC gene phylogenetic tree genetic evolution analysis showed that the phylogenetic tree is divided into two branches,one of which is from the domestic classic strain.The Chinese strains are occupied by two small branches of the mutant strains isolated after 2011.the other is the European and American strains composed of European isolates and American isolates.The results of micro-intersection neutralization test showed that the titers of PRV-Bartha K61 high serum-free neutralizing PRV-GD1406 and PRV-Bartha K61 were 22 and 53,respectively,and PRV-GD1406 high serum-free neutralizing PRV-GD1406 and PRV-Bartha K61 was 35.68 and 44 respectively;the heterologous serum neutralizingtiter/homologous serum neutralizing titer of PRV-GD1406 strain was≈0.83,and the PRV-Bartha K61 strain heterologous serum neutralizing titer/homologous serum neutralization titer was≈0.61;the correlation coefficient of cross-reactivity between the two strains(PRV-GD1406,PRV-Bartha K61)was R=72.This study can provide theoretical basis and new ideas for the prevention and control of pseudorabies work and screening vaccine strains.
