摘  要:目的优化破壁灵芝孢子粉和CO_2超临界流体萃取(SFECO_2)孢子油后破壁灵芝孢子粉中水溶性多糖提取技术。方法本课题以项目前期已确定的最优三萜提取技术为基础,设计四因素三水平正交实验表。以水为溶剂,回流法提取两类破壁灵芝孢子粉残渣。根据药典,使用蒽酮-硫酸法测定吸光度,计算多糖含量,确定最佳提取方案。结果在前期各自最优三萜提取方法下,灵芝破壁孢子粉水溶性多糖最优提取方法是:温度70℃,液料比30∶1(mL·g^(-1)),时间2h,提取次数1次;SFECO_2孢子油后破壁灵芝孢子粉水溶性多糖最优提取方法是:温度90℃,液料比40∶1(mL·g^(-1)),提取时间1.5h,提取次数2次。结论确定了两类灵芝破壁孢子粉最优提取方法,进一步提高对破壁灵芝孢子粉的利用,为其综合开发利用提供系统、全面的依据。OBJECTIVE To optimize the extraction technology of WSP from Wall-broken Ganoderma lucidum spore powder and Wall-broken Ganoderma lucidum spore powder after SFECO 2.METHODS Based on the optimal triterpene extraction technology determined in the early stage of the project.This subject designed a four-factor and three-level orthogonal experiment table.Using water as solvent,the reflux method was used to extract two kinds of Wall-broken Ganoderma lucidum spore powder residue.According to the pharmacopoeia,anthrone-sulfuric acid methodwas used to determine absorbance,calculate the content of polysaccharides,and determine the optimal extraction scheme.RESULTS Under the previousoptimal triterpenoid extraction method,the optimal extraction method ofWSP from Wall-broken Ganoderma lucidum spore powder was:the temperature was 70℃,the liquid to material ratio was 30∶1(mL·g-1),the time was 2h and the extraction times was one time.And the optimal extraction method of WSP from Wall-broken Ganoderma lucidum spore powder after SFECO 2 was:the temperature was 90℃,the liquid to material ratio was 40∶1(mL·g-1),the time was 1.5h and the extraction times was two times.CONCLUSION The optimal extraction methods for two types of Ganoderma lucidum spore powder were determined to further improvethe utilization of Ganoderma lucidum spore powder,and provide a systematic and comprehensive basis for its comprehensive development and utilization.
