裕丹参对实验性心肌梗死大鼠心肌组织的保护作用

摘  要:目的探讨裕丹参对实验性心肌梗死大鼠的保护作用及其机制。方法随机将Wistar大鼠50只分为假手术组、模型组和低、中、高剂量裕丹参组。后三组分别以15、30、60 mg/kg/d的剂量灌胃给药,前两组相同量的生理盐水灌胃。4周后检测各组大鼠的血液动力学参数,取颈动脉血液,测定血清中乳酸脱氢酶(LDH)、肌酸激酶-MB(CK-MB)、前列环素(PGI2)和血栓素A2(TXA2)的含量。处死大鼠,取心尖部心肌组织,一半做石蜡切片,HE染色检测心肌组织的病理变化,TUNEL免疫荧光染色检测心肌细胞的凋亡数量变化,免疫组化法检测Caspase-3蛋白的表达水平;另一半制作成组织匀浆,检测心肌组织中的超氧化物歧化酶(SOD)和丙二醛(MDA)的含量及Caspase-3蛋白、B细胞淋巴瘤-2蛋白(Bcl-2)和Bcl-2关联蛋白(Bax)的表达水平。结果血流动力学检测结果证实,各剂量裕丹参治疗组均可明显升高大鼠心脏的左室收缩压、左室内压曲线最大上升和下降速率,降低左心室舒张末期压力。血清酶学结果表明,各剂量裕丹参治疗组均可明显升高血清中的PGI2浓度,降低血清中的LDH、CK-MB、TXA2浓度。HE染色、TUNEL染色和免疫组化染色结果证实,各剂量裕丹参治疗组均可明显改善心肌组织的紊乱形态,显著减少凋亡的心肌细胞数量,下调心肌组织中的Caspase-3蛋白表达。心肌组织检测结果表明,各剂量裕丹参治疗组均可明显上调心肌组织中的SOD含量和Bcl-2蛋白的表达,下调组织中的MDA含量、Caspase-3和Bax蛋白的表达。结论裕丹参对实验性心肌梗死大鼠的心肌具有明显的保护作用,其作用可能与裕丹参具有抗氧化、抗凋亡的作用密切相关。Objective In order to investigate the protective effect of Yu Danshen on experimental rats with myocardial infarction and its mechanism.Methods 50 Wistar rats were randomly divided into sham operation group,model group and low,middle and high dose Yudanshen group.The latter three groups were given Yudanshen by intragastric administration at doses of 15,30,60 mg/kg/d,respectively.The former two groups were given the same amount of saline.Four weeks later,the hemodynamic parameters of rats in each group were measured.The contents of lactate dehydrogenase(LDH),creatine kinase-MB(CK-MB),prostacyclin(PGI2) and thromboxane A2(TXA2) in serum were measured.The rats were sacrificed,half of the apical myocardium was taken for paraffin sections.The pathological changes of the myocardium were detected by HE staining,the apoptosis of myocardial cells was detected by TUNEL immunofluorescence staining,and the expression of Caspase-3 protein was detected by immunohistochemistry.The other half was made into tissue homogenate to detect the content of superoxide dismutase(SOD),malondialdehyde(MDA) in myocardial tissue and the expression of Caspase-3 protein,B-cell lymphoma-2 protein(Bcl-2) and Bcl-2 associated protein(Bax).Results Yudanshen could significantly increase the left ventricular systolic pressure,the maximum increase and decrease rate of left ventricular pressure curve,and decrease the left ventricular end-diastolic pressure.The results of serum enzymology showed that the levels of PGI2 were significantly increased,while the levels of LDH,CK-MB and TXA2 were decreased in serum of Yudanshen treatment group.HE staining,TUNEL staining and immunohistochemical staining showed that Yudanshen could improve the disorder of myocardial tissue,reduce the number of apoptotic myocardial cells,and down-regulate the expression of Caspase-3 protein in myocardial tissue.The results of myocardial tissue test showed that compared with the model group,the SOD content and Bcl-2 protein expression in myocardium were significantly increased an
