摘 要:开花抑制基因FLC(FLOWERING LOCUS C)是春化作用中的关键基因,为探索MADS-box转录因子在芒果花器官发育中的作用,本研究利用RT-PCR克隆得到芒果MADS-box转录因子MiFLC的cDNA全长,序列分析显示该基因具有MADS_MEF2_like和K-box结构域,属于MADS-box基因家族,MiFLC基因编码区长576 bp,编码191个氨基酸,蛋白质相对分子质量22.08 kD,等电点为9.18。MiFLC的生物信息学分析表明该蛋白序列的二级结构主要由α螺旋、无规则转曲及延伸链构成,属于亲水性氨基酸。序列分析和系统进化树分析显示,MiFLC基因编码的蛋白与橙子、枳、龙眼、克莱门柚的蛋白序列同源性高,亲缘关系较近。组织特异性表达分析表明,MiFLC基因在芒果不同品种和每个品种的不同组织部位均有表达,在‘桂七’、‘金煌’、‘凯特’3个品种的表达量呈现出一致性,在营养器官中表达量最高,在花器官中表达量较低,表达量顺序依次是叶>茎>果>花。FLC(FLOWERING LOCUS C)is a key gene in the vernalization.To explore the role of MADS-box transcription factor in the development of mango flower organs,this study used RT-PCR to clone the mango MADS-box transcription factor Mi FLC.The full-length cDNA,sequence analysis showed that the gene has MADS_MEF2_like and K-box domains,belonging to MADS-box gene family.The MiFLC gene coding region is576 bp long and encodes 191 amino acids.The relative molecular mass of the protein is 22.08 kD and the isoelectric point is 9.18.Bioinformatics analysis of MiFLC showed that the secondary structure of the protein sequence is mainly composed of alpha helix,random rotation and extended chain,and belong to hydrophilic amino acids.Sequence analysis and phylogenetic tree analysis showed that the protein encoded by MiFLC gene had high homology with the protein sequences of Citrus sinensis,Citrus trifoliata,Dimocarpus longan Lour.and citrus clementina,and the genetic relationship was close.The tissue-specific expression analysis indicated that the MiFLC gene was expressed in different varieties of mango and in different tissues of each variety.The expression levels of the three varieties of’Guiqi’,’Jinhuang’and’Keitt’showed consistency and expression in vegetative organs.The highest,the expression level in the flower organs is lower,and the order of expression is leaf>stem>fruit>flower.
