摘 要:针对银杏幼茎尖在组培过程中容易出现的褐化问题,从外植体的幼嫩程度、选取长度、培养基的pH值、培养温度以及防褐剂的类型对银杏幼茎尖褐化的影响来控制银杏幼茎尖的褐化率。结果表明,防褐剂对银杏幼茎尖的褐化率的影响不大;选择当年生嫩枝条上2~3 cm的幼茎尖作为外植体,预培养温度15℃~20℃,培养基pH值为6~8时能将银杏幼茎尖的褐化率有效地控制在10%以下。通过探讨不同培养条件对降低银杏幼茎尖组织培养褐化率的效果,有利于促进银杏幼茎尖的丛生芽和根的诱导,达到优化银杏幼茎尖组织培养条件的目的。In order to reduce the browning rate of young stem tip of Ginkgo biloba by studying on the influence of the tender degree of explants,the length of explants,the pH value of the medium,the culture temperature and the type of anti-browning agent.The results showed that the addition of different kinds of anti-browning agents had little effect on the browning rate of young stem tip.The browning rate of young stem tip of Ginkgo biloba could be effectively controlled below 10%,when the annual shoot stem tip were selected as explants,pre-cultured at 15℃~20℃medium,and pH at 6~8.By discussing the influence of different cultural conditions on reducing browning rate of tissue culture of young stem tip of Ginkgo biloba,which was beneficial to promote induction of cluster buds and roots,and optimize tissue culture conditions of young stem tip of Ginkgo biloba.
