高效液相色谱法测定阿比特龙的纯度

摘　　要：建立测定阿比特龙纯度的高效液相色谱法。色谱条件如下:色谱柱为Agilent C18柱(250 mm×4.6 mm,3.5μm),流动相为乙腈-水(1∶1),流量为1.2 mL/min,检测波长为254 nm,柱温为25℃,进样体积为20μL。阿比特龙的质量浓度在40~520μg/mL范围内与色谱峰面积线性关系良好,线性方程为A=31.9572c+136.1652,相关系数为0.9997,检出限为0.24μg/mL。样品测定结果的相对标准偏差为0.27%(n=6),标准样品回收率为99.12%~99.88%。该法专属性强,准确、可靠,具有较高的灵敏度和精密度。A method of high performance liquid chromatography(HPLC)for the determination of abiraterone was established.The chromatographic conditions were as follows:Agilent C18 column(250 mm×4.6 mm,3.5μm),acetonitrile-water solution(1∶1)as the mobile phase with the flow rate of 1.2 mL/min,detection wavelength of 254 nm,column temperature of 25℃,and injection volume of 20μL.The mass concentration of abiraterone was proportional to the chromatographic peak area in the range from 40 to 520μg/mL.The linear regression equation was formulated as:A=31.9572c+136.1652 with the correlation coefficient of 0.9997.The detection limit was 0.24μg/mL.The relative standard deviation of the detection results was 0.27%(n=6).The recoveries of standard samples were in the range from 99.12%to 99.88%.The method is specific,accurate and reliable with high sensitivity and precision.
