摘 要:采用RT-PCR的方法对贵州黑山羊LYRM1基因进行了克隆,并对该基因的编码区序列进行生物信息学分析,利用qRT-PCR的方法检测LYRM1基因在贵州黑山羊不同组织中mRNA的相对表达量以及在不同月龄的时序表达。结果:黑山羊LYRM1基因的编码区全长369 bp,编码122个氨基酸,编码蛋白分子质量为14.4 kDa,理论等电点为9.73,不稳定系数为49.48,为不稳定蛋白;与GenBank数据库中提供的山羊LYRM1基因的编码区序列匹配度为99%,存在2个碱基的突变,分别为第177位和242位;通过LYRM1基因系统进化树得知,黑山羊与牛的进化距离较近,与斑马鱼进化距离较远。qRT-PCR结果显示LYRM1基因在贵州黑山羊7个组织中均有表达,在脂肪组织的表达量最高,背最长肌中的表达量最低;时序表达结果表明,LYRM1基因在黑山羊脂肪组织的整体表达趋势为先增加再减少,6月龄的表达量最高,7日龄最低。本试验成功克隆了黑山羊LYRM1基因编码区并进行了生物信息学分析,且检测了其在黑山羊不同组织中的表达以及在脂肪组织中的时序表达,为深入研究LYRM1基因功能奠定基础。The LYRM1 gene of black goat was cloned using RT-PCR,and the coding region sequence of the gene was analyzed by bioinformatics.The relative expressions of the LYRM1 gene in different tissues of Guizhou black goat and its expression sequences at different months of age were detected using qRT-PCR.The results showed that the coding region of LYRM1 in the black goat was 369 bp,encoding 122 amino acids.Its protein molecular weight was 14.4 kDa,its theoretical isoelectric point was 9.73,and its instability point coefficient was 49.48;which indicated an unstable protein.The sequence matching degree of the coding region of the gene provided in GenBank database was 99%,and it had two base mutations,at positions of 177 and 242,respectively.The phylogenetic tree of LYRM1 indicatred that the evolutionary distance between black goats and cattle was relatively close,while that between black goats and zebrafish was relatively distant.The qRT-PCR results showed that LYRM1 was expressed in 7 tissues of the Guizhou black goat,with the highest expression in the adipose tissue and the lowest expression in the dorsi longus muscle.The sequence expression results showed that the overall expression trend of LYRM1 in the fat tissues of the black goats first increased and then decreased,with the highest expression at 6 months of age and the lowest expression at 7 days of age.In this experiment,the coding region of the LYRM1 gene in black goat was cloned successfully and bioinformatics analysis was carried out and its expression in different tissues of black goat and expression sequence in adipose tissue were detected;which laid a foundation for further study on the function of the LYRM1 gene.
