摘  要:目的研究冷刺激对大鼠精子功能的影响,并建立冷刺激诱导的弱精子症大鼠模型,探讨其性腺轴功能改变。方法取16只雄性SD大鼠在4℃低温环境(8 h/d)建模,根据弱精子症标准筛选并建立弱精子症模型组(8只)及未成模组(8只);另取12只SD大鼠为正常对照组。饲养23周后取材称睾丸与附睾质量计算器官系数,左侧附睾用于检测各组大鼠精子浓度、精子活力及活率,并对模型予以综合评价。利用放射免疫试剂盒检测各组血清T、E2、LH、FSH、PRL激素水平,HE染色后光镜下观察各组大鼠下丘脑、垂体、睾丸、附睾形态学改变。结果与正常对照组相比,冷刺激建模组、弱精子症组大鼠精子活力、活率降低,差异有统计学意义(P<0.05);弱精子症组大鼠睾丸、附睾系数降低,差异有统计学意义(P<0.01);弱精子症组大鼠E2水平升高,LH水平降低,差异有统计学意义(P<0.05)。光镜下可见正常对照组大鼠下丘脑、垂体、睾丸和附睾结构正常,未成模组、弱精子症组大鼠下丘脑、垂体、睾丸和附睾结构均有不同程度的病理改变。结论冷刺激可降低大鼠精子活力和活率,冷刺激可诱导建立弱精子症大鼠模型,其机制与性腺轴功能改变有关。Objective To study the effect of cold stimulation on sperm function in rats,to establish a rat model of asthenospermia induced by cold stimulation,and to explore the changes of gonadal axis function.Methods Sixteen male Sprague Dawley(SD)rats were set up the model at 4℃for 8 h/d.According to the criteria of asthenospermia,8 rats were selected as asthenospermia model group,and 12 SD rats were selected as normal control group.After 23 weeks of feeding,the weights of testis and epididymis were measured to calculate the organ coefficient.The left epididymis was used to detect sperm concentration,sperm motility and motility rate in each group.The serum levels of LH,FSH,and hypothalamus were detected by immunocytochemistry.Results Compared with the normal control group,the sperm motility and viability of rats in cold stimulation model group and asthenospermia group decreased significantly(P<0.05).The coefficients of testis and epididymis in asthenospermia group were significantly lower than those in the normal control group(P<0.01).Compared with the normal control group,the level of E2 in the asthenospermia group was higher,while the level of LH was lower in the asthenospermia group.Byoptical microscope,the hypothalamus,pituitary,and testicular structures of the normal control group were normal,and the vary changes of pathological wereobviousin the unformed and asthenospermia groups.Conclusion Cold stimulation can significantly reduce the sperm motility and viability of rats,which can induce the establishment of asthenospermia rat model,the mechanism may be related to the changes of gonadal axis function.
