摘 要:目的探讨七叶皂苷钠对氧化应激诱导的大鼠晶状体组织损伤的抑制作用及其机制。方法分别以不同时限观察空白对照组、七叶皂苷钠组、氧化损伤组、氧化损伤+七叶皂苷钠组的大鼠晶状体混浊情况,应用HE染色及电镜进行组织学观察,ELISA法检测晶状体组织匀浆中还原性谷胱甘肽(GSH)含量及过氧化氢酶(CAT)活力,Western-blot法观察晶状体上皮细胞凋亡变化。结果(1)晶状体透明度观察,氧化损伤组晶状体透明度明显下降,而氧化损伤+七叶皂苷钠组晶状体混浊情况明显轻于氧化损伤组;(2)HE染色结果显示,空白对照组及七叶皂苷钠组晶状体上皮细胞为单层立方形细胞,排列整齐,而氧化损伤组晶状体边缘不规则,晶状体上皮细胞排列紊乱,给予七叶皂苷钠干预后,细胞形态逐渐规则;(3)电镜观察结果显示,氧化损伤组晶状体上皮细胞基质密度减低,胞质内空泡形成,细胞核肿胀,给予七叶皂苷钠干预后,大鼠晶状体上皮细胞形态明显改善;(4)与氧化损伤组相比,经七叶皂苷钠治疗后GSH含量及CAT活力表达量均升高,tGSH=5.573,PGSH=0.005;tCAT=4.118,PCAT=0.015,均有统计学意义;(5)Western-blot结果显示,氧化损伤+七叶皂苷钠组大鼠晶状体中NF-κB的蛋白表达量降低,与氧化损伤组比较差异有统计学意义(t=3.571,P=0.023)。结论七叶皂苷钠可抑制氧化损伤所诱导的大鼠晶状体组织损伤并延缓白内障形成,有望成为治疗白内障的有效药物。OBJECTIVE To investigate the inhibitory effect and its mechanism of sodium aescinate on oxidative stress-induced rat lens injury.METHODS The rat lens opacification of blank control group,sodium aescinate group,oxidative damage and oxidative damage+sodium aescinate group were observed in different time limit respectively.HE staining and electron microscopy were applied for histological observation.The content of reduced glutathione(GSH)and the activity of catalase(CAT)in lens tissue homogenate were detected by ELISA.The apoptosis of lens epithelial cells was observed by Western blot.RESULTS Firstly,the results of lens transparency observation showed that the lens transparency of the oxidative damage group were decreased obviously,while the lens opacity of the oxidative damage+aescinate group were obviously lighter than that in oxidative damage group.Secondly,HE staining results showed that lens epithelial cells of the blank control group and sodium aescinate group were monolayer cubic cells,which were rowed neatly,while the lens edge of the oxidative damage group was irregular,and the lens epithelial cells were arranged disorderly.After the intervention of sodium aescinate,the cell morphology gradually became regular.Thirdly,the results of electron microscopy showed that the matrix density of lens epithelial cells of the oxidative damage group were decreased,vacuoles formed in the cytoplasm and nuclei swelled.After the intervention of sodium aescinate,the morphology of lens epithelial cells in rats was improved significantly.Fourthly,compared with the oxidative damage group,GSH content and CAT activity expression of oxidative damage+aescinat group were increased after treatment with sodium aescinate(tGSH=5.573,PGSH=0.005;tCAT=4.118,PCAT=0.015).Lastly,the results of Western blot showed that the protein expression of NF-κB in the lens of oxidative damage+aescinat group was significantly decreased,compared with the oxidative damage group,and the differences were statistically significant(t=3.571,P=0.023).CONCLUSIONS
