摘 要:目的观察藻蓝蛋白对大鼠心肌缺血-再灌注损伤的影响并分析可能机制。方法选取大鼠36只,用随机数字表法均分为空白对照组、模型组和治疗组。模型组和治疗组采用反复3次丝线结扎、复灌左冠状动脉前降支45 min的方法建模。建模后治疗组用10%藻蓝蛋白混悬液以200 mg/(kg·d)分2次灌胃治疗7 d,空白对照组和模型组用等体积生理盐水灌胃。采用图像分析法检测大鼠冠状动脉细胞周期蛋白依赖激酶抑制因子p27、p21表达;采用免疫组织化学染色后检测诱导型一氧化氮合酶(iNOS);酶联免疫吸附法检测大鼠心室内动脉血清超氧化物歧化酶(SOD)、肌酸激酶同工酶(CK-MB)、丙二醛(MDA)和一氧化氮(NO)。结果治疗组大鼠冠状动脉细胞周期蛋白依赖激酶抑制因子p27、p21高表达,iNOS阳性细胞及SOD、NO水平升高,CK-MB、MDA水平均明显降低,与模型组比较,差异均有统计学意义(P<0.05)。结论藻蓝蛋白具有抑制心肌缺血-再灌注损伤的效应。Objective To observe the effect of phycocyanin on myocardial ischemia-reperfusion injury in rats and analyze the possible mechanism.Methods Thirty-six rats were selected and divided into blank control group,model group and treatment group by random number table method.Model group and treatment group were modeled by repeating 3 times of silk thread ligation and reperfusion of the left anterior descending coronary artery for 45 minutes.After modeling,the treatment group was treated with 10%phycocyanin suspension at 200 mg/(kg·d)twice by gavage for 7 days,and the blank control group and model group were treated with equal volume of saline.Image analysis method was used to detect the expression of cyclin-dependent kinase inhibitor p27 and p21 in rats coronary artery;immunohistochemical staining was used to detect inducible nitric oxide synthase(iNOS);enzyme-linked immunosorbent assay was used to detect rats ventricular arterial serum superoxide dismutase(SOD),creatine kinase isoenzyme(CK-MB),malondialdehyde(MDA)and nitric oxide(NO)in rats.Results In the treatment group,the coronary artery cyclin-dependent kinase inhibitors p27 and p21 were highly expressed,the levels of iNOS positive cells,SOD and NO were increased,and the levels of CK-MB and MDA were significantly reduced.Compared with the model group,the difference was statistically significant(P<0.05).Conclusion Phycocyanin has the effect on inhibiting myocardial ischemia-reperfusion injury.
