摘 要:目的采用液相色谱-串联质谱法与酶联免疫法测定人血浆地高辛浓度并对两种方法比较,探讨人血浆地高辛测定优化方法。方法建立液质联用技术(high performance liquid chromatography-tandem mass spectrometry,HPLC-MS/MS)法并对其方法学进行考察,分别在独立的实验室用HPLC-MS/MS法和酶放大免疫(enzyme-multiplied immunoassay technique,EMIT)法检测质控样品和37例临床生物样本血浆中地高辛的浓度,并比较两种方法的优劣性。结果 37例临床患者血浆样本测定结果显示,EMIT法的测定结果优于HPLCMS/MS法(P<0.05)。结论 HPLC-MS/MS法比EMIT法在测定人血浆地高辛血药浓度时特异性更强,更能准确反映真实血浆药物浓度。Objective To compare and optimize the methods of HPLC-MS/MS and EMIT for detemination of digoxin concentration in human plasma. Methods Aim to establish an HPLC-MS/MS method for the assay of digoxin.The plasma digoxin concentration of 37 clinical biological samples were detected by HPLC-MS/MS and EMIT respectively in independent laboratories. The results of the two methods were compared by paired t-test for statistical test. Result The results in samples of EMIT were higher than those of HPLC-MS/MS. There were statistical differences between the two methods. Conclusion HPLC-MS/MS method is more specific than EMIT method in the determination of digoxin in human plasma. It can reflect the real plasma concentration more accurately.
