摘  要:目的:探讨白桦脂醇对人胃癌MGC-803细胞增殖和凋亡的影响。方法:CCK-8试剂盒检测不同浓度白桦脂醇对细胞活性的影响;Western blot检测细胞中Ki67和增殖细胞核抗原(PCNA)表达水平;流式细胞术分析细胞凋亡情况;Western blot检测细胞中凋亡相关蛋白和磷脂酰肌醇3激酶(PI3K)/磷酸化蛋白激酶B(AKT)通路相关蛋白表达水平。结果:白桦脂醇浓度低于50μmol/L时,对细胞活性没有显著影响;不同浓度白桦脂醇处理人胃癌MGC-803细胞后,细胞增殖倍数显著降低;细胞中Ki67和PCNA的表达水平显著降低;发生凋亡的细胞比率显著增加,细胞中Bcl-2表达水平显著降低,Bax、Cleaved caspase-9和Cleaved caspase-3表达水平显著升高;细胞中p-PI3K/PI3K、p-AKT/AKT比率显著降低,p65和mTOR表达水平显著降低,上述作用均具有白桦脂醇剂量依赖效应。结论:白桦脂醇抑制胃癌MGC-803细胞增殖,诱导细胞凋亡,其作用机制可能与抑制PI3K/AKT通路激活相关。Objective:To investigate effects of betulin on proliferation and apoptosis of human gastric cancer MGC-803 cells.Methods:CCK-8 kit was used to detect effect of cell viability with different concentrations of betulin.Expression levels of Ki67 and proliferating cell nuclear antigen were detected by Western blot;flow cytometry was used to detect cell apoptosis rate;expression levels of apoptosis-related proteins and PI3K/AKT pathway related proteins were perfornde by Western blot.Results:Betulin had no significant effect on cell viability when its concentration was lower than 50μmol/L;after treated with different concentrations of betulin,human gastric cancer MGC-803 cells proliferation rate decreased significantly,expression levels of Ki67 and PCNA decreased significantly;apoptotic cell ratio increased significantly,expression levels of Bcl-2 in cells decreased significantly,and expression levels of Bax,Cleaved-caspase-3 and Cleaved-caspase-9 in human gastric cancer MGC-803 cells increased significantly;ratio of p-PI3K/PI3K and p-AKT/AKT in cells were decreased significantly,and expression levels of p65 and mTOR decreased significantly.The above effects were dose-dependent.Conclusion:Betulin inhibits proliferation and induces apoptosis of gastric cancer MGC-803 cells,and its mechanism may be related to inhibition of PI3K/AKT pathway activation.
