益气活血药对心梗后心肌纤维化大鼠不同亚型血小板源性生长因子表达的影响

摘　　要：目的探究益气活血药对心肌梗死后心肌纤维化大鼠血小板源性生长因子(platelet-derived growth factors,PDGF)表达的影响。方法结扎左前降支建立心肌梗死大鼠模型,假手术组只穿线,不结扎。术后将成模动物随机分为假手术组、模型组、培哚普利组和益气活血组,每组9只,给予相应药物灌胃治疗28天后,采用超声心动图评价大鼠心脏结构和功能的变化,马松(masson)染色法观察药物对心肌纤维化的影响,蛋白质印迹法(western blot)检测collagenⅠ、collagenⅢ及PDGFs的蛋白表达水平。结果与假手术组比较,模型组大鼠左室射血分数(left ventricular ejection fraction,LVEF)、左室短轴率(left ventricular fraction shortened,LVFS)均显著降低(P<0.01),左室收缩末期内径(left ventricular internal dimension systole,LVIDs)、左室舒张末期内径(left ventricular end diastolic diameter,LVIDd)、心肌胶原容积分数(collagen volume fraction,CVF)显著升高(P<0.01),collagenⅠ、Ⅰ/Ⅲ型胶原比值和PDGF-B升高(P<0.05,P<0.01),PDGF-A和PDGF-C显著下降(P<0.01)。与模型组相比,两给药组LVEF、LVFS均显著升高(P<0.01),LVIDs、LVIDd、CVF显著降低(P<0.01),其中益气活血组collagenⅠ、Ⅰ/Ⅲ型胶原比值和PDGF-B下降(P<0.01),PDGF-A升高(P<0.05)。结论益气活血药作用于血小板源性生长因子能够通过降低PDGF-B减轻MI大鼠纤维化水平,并可上调PDGF-A发挥保护心脏功能的作用。Objective To explore the effects of drugs for Yiqi Huoxue on expression of platelet-derived growth factor(PDGF)in myocardial fibrosis rats after myocardial infarction.Methods The rat model with myocardial infarction was established by ligating the left anterior descending branch.In the sham group,thread was threaded without ligation.After operation,the model rats were randomly divided into sham operation group,model group,perindopril group,and Yiqi Huoxue group,with 9 rats in each group.Afert giving gavage treatment with corresponding drug for 28 days,echocardiography was used to evaluate the changes of heart structure and function in rats,Masson staining was used to observe the effects of drugs on myocardial fibrosis,Western blot was used to detect the expression levels of collagenⅠ,collagenⅢ,and PDGFs.Results Compared with the sham operation group,the left ventricular ejection fraction(LVEF)and left ventricular fraction shortened(LVFS)of the rats in the model group were significantly reduced(P<0.01).Left ventricular internal dimension systole(LVIDs),left ventricular end diastolic diameter(LVIDd)and myocardial collagen volume fraction(CVF)significantly increased(P<0.01),collagenⅠ,Ⅰ/Ⅲcollagen ratio and PDGF-B increased and there was significant different(P<0.05,P<0.01),PDGF-A and PDGF-C decreased significantly(P<0.01).Compared with the model group,LVEF and LVFS in perindopril group and Yiqi Huoxue group were significantly increased(P<0.01),LVIDs,LVIDd and CVF were significantly reduced(P<0.01).whileⅠ/Ⅲcollagen ratio and PDGF-B decreased(P<0.01),and PDGF-A increased(P<0.05).Conclusion Effects of drugs for Yiqi Huoxue on platelet-derived growth factor can reduced the fibrosis levels of MI rats by reducing PDGF-B,protecting the heart function upregulating PDGF-A.
