摘 要:目的:基于miR-155/SOCS1轴研究复方甘草酸苷治疗特应性皮炎(AD)小鼠的作用机制。方法:BALB/c小鼠随机分为对照组、模型组、miR-155 agomir组、miR-155 agomir阴性对照组、复方甘草酸苷组、复方甘草酸苷+miR-155 agomir组。除对照组外,其余各组小鼠采用耳背涂布卡泊三醇(MC903)溶液法建立AD小鼠模型。观察各组小鼠耳部临床特征并进行皮炎评分;HE检测各组小鼠耳组织病理情况;ELISA检测各组小鼠血清肿瘤坏死因子α(TNF-α)、IL-6、IgE水平,qRT-PCR法检测各组小鼠耳部出现组织中miR-155、SOCS1 mRNA水平;Western blot检测各组小鼠耳部组织中SOCS1蛋白表达。结果:与对照组比,模型组小鼠耳部出现明显红斑、充血红肿、耳组织表皮层增生、棘细胞层变厚、毛细血管扩张、组织水肿充血、大量炎症细胞浸润等症状,耳部皮炎评分、血清IgE及TNF-α、IL-6水平、组织miR-155水平升高(P<0.05),SOCS1 mRNA及蛋白水平降低(P<0.05)。与模型组相比,miR-155 agomir组小鼠上述症状加重,耳部皮炎评分、血清IgE及TNF-α、IL-6水平、组织miR-155水平升高(P<0.05),SOCS1 mRNA和蛋白表达降低(P<0.05);miR-155 agomir阴性对照组小鼠各指标差异无统计学意义(P>0.05);复方甘草酸苷组小鼠上述症状减轻,耳部皮炎评分、血清IgE及TNF-α、IL-6水平、组织miR-155水平降低(P<0.05),SOCS1 mRNA和蛋白表达升高(P<0.05)。与miR-155 agomir组相比,复方甘草酸苷+miR-155 agomir组小鼠上述症状减轻,耳部皮炎评分、血清IgE及TNF-α、IL-6水平、组织miR-155水平降低(P<0.05),SOCS1 mRNA和蛋白表达升高(P<0.05)。与复方甘草酸苷组相比,复方甘草酸苷+miR-155 agomir组小鼠上述病理症状加重,耳部皮炎评分、血清IgE及TNF-α、IL-6水平、组织miR-155水平升高(P<0.05),SOCS1 mRNA和蛋白表达降低(P<0.05)。结论:复方甘草酸苷可通过下调miR-155表达、上调SOCS1表达减轻AD小鼠炎症反应。Objective:To study mechanism of compound glycyrrhizin in treatment of atopic dermatitis(AD)mice based on miR-155/SOCS1 axis.Methods:BALB/c mice were randomly divided into control group,model group,miR-155 agomir group,miR-155 agomir negative control group,compound glycyrrhizin group,compound glycyrrhizin+miR-155 agomir group.Except for control group,AD mice in other groups were established by using method of ear back coating with captopril(MC903)solution.Clinical features of ears of mice in each group were observed and dermatitis scores were scored,pathological changes of ear tissues of mice in each group were detected by HE,levels of serum tumor necrosis factor-α(TNF-α),IL-6 and IgE were detected by ELISA,qRT-PCR was used to detect levels of miR-155 and SOCS1 mRNA in ear tissues of mice in each group,and expression of SOCS1 protein in ear tissues of mice in each group was detected by Western blot.Results:Compared with control group,ears of mice in model group had obvious erythema,hyperemia and swelling,epidermal layer proliferation of ear tissues,thickening of spinous cell layer,telangiectasia,edema and hyperemia,and a large number of inflammatory cell infiltration.Ear dermatitis scores,serum IgE and TNF-α,IL-6 levels,tissue miR-155 level were increased(P<0.05),and expression levels of SOCS1 mRNA and protein were decreased(P<0.05).Compared with model group,the above-mentioned symptoms were aggravated in miR-155 agomir group,ear dermatitis scores,serum IgE and TNF-α,IL-6 levels,miR-155 level in tissues were increased(P<0.05),and expression levels of SOCS1 mRNA and protein were decreased(P<0.05).While there was no significant difference in indices of mice in control group(P>0.05).In compound glycyrrhizin group,the above symptoms were alleviated,ear dermatitis scores,serum IgE and TNF-α,IL-6 levels,tissue miR-155 level were decreased(P<0.05),and expression levels of SOCS1 mRNA and protein were increased(P<0.05).Compared with miR-155 agomir group,the above-mentioned symptoms of mice in compound glycyrrhizin+miR
