摘 要:[目的]建立能够有效提高蝉拟青霉液体培养效率的培养体系,增加其生物量。[方法]以蝉拟青霉生物量作为响应值,通过Plackett-Burman实验设计从8个试验影响因素中筛选出3个主要影响因素,基于3因素3水平的Box-Behnken设计法以及响应面分析法研究影响蝉拟青霉液体培养各因素之间的交互作用。[结果]通过Plackett-Burman设计法确定影响最为显著的因素为接种量、pH和温度,利用最陡爬坡试验确定最佳范围在接种量7%、pH 6.00、温度25℃附近,借助Box-Behnken设计法最终确定最佳液体培养条件为:葡萄糖20 g/L、蛋白胨3 g/L、酵母膏2 g/L、KH2PO41 g/L、MgSO40.5 g/L、接种量6.65%、pH 6.00、温度24.01℃。优化后的培养体系所培养的蝉拟青霉其生物量从优化前的147.1060 g/L提高到239.4753 g/L,提高了62.79%。[结论]优化后的培养体系能够有效提高蝉拟青霉生物量,该方法对于优化蝉拟青霉生物量发酵条件是一种有效、可行的方法。[Objective]To establish a culture system that can effectively improve the liquid culture efficiency of Paecilomyces cicadae and increase its biomass. [Method]Using Paecilomyces cicada biomass as the response value,three main influencing factors were selected from the eight experimental influencing factors through the Plackett-Burman experimental design,and then the Box-Behnken design method with 3 factors and 3 levels and response surface analytical methods were used to study the interaction between various factors affecting the liquid culture of Paecilomyces cicadae.[Result]The Plackett-Burman design method was used to determine the most influential factors were inoculation volume,pH and temperature. The steepest climbing test was used to determine the optimal range to be around 7% inoculation volume,pH 6. 00 and temperature 25 ℃ .Box-Behnken design method finally determined the best liquid culture conditions: glucose 20 g/L,peptone 3 g/L,yeast extract 2 g/L,KH_2PO_4 1 g/L,MgSO_4 0. 5 g/L,inoculation amount 6. 65%,pH 6. 00,temperature 24. 01 ℃ . The biomass of Paecilomyces cicadae cultured by the optimized medium increased from 147. 106 0 g/L before optimization to 239. 475 3 g/L,an increase of 62. 79%.[Conclusion] The optimized culture system can effectively improve the biomass of Paecilomyces cicadae,indicating that this method is a feasible method for optimizing the fermentation conditions of Paecilomyces cicadae biomass.
