摘 要:目的:对6个柴胡种质的抗寒性及其与游离脯氨酸、可溶性蛋白、可溶性糖含量等生理指标进行相关性分析,为柴胡种质的抗寒性评价、优良种苗筛选和人工栽培提供理论依据。方法:分别采集甘肃定西、山东济南、湖北十堰、河北涉县、山西万荣、陕西陇西共6份种子,编号依次为GS-dx、SD-jn、HB-sy、HB-sx、SX-wr、SX-lx。于柴胡越冬期、返青期进行生长状况及死苗率调查;茚三酮比色法测定游离脯氨酸含量,考马斯亮蓝G-250染色法测定可溶性蛋白含量,蒽酮比色法测定可溶性糖含量。结果:6个柴胡种质的越冬死苗率差异显著,HB-sx的死苗率最低为7.81%,其次为GS-dx的12.87%,死苗率依次为SX-lx>SX-wr>HB-sy>SD-jn>GS-dx>HB-sx。越冬期和返青期,6个柴胡种质根系和叶片的游离脯氨酸、可溶性蛋白及可溶性糖含量均差异显著,越冬期GS-dx根系的游离脯氨酸含量最高为1.35 mg/g,HB-sx根系的可溶性蛋白含量1.37μg/g和可溶性糖含量3.88%均最高;HB-sx叶片的游离脯氨酸含量最高为0.58 mg/g,GS-dx叶片的可溶性蛋白含量0.97μg/g和可溶性糖含量3.48%均最高。柴胡越冬期根系、叶片的游离脯氨酸含量与越冬死苗率均呈显著负相关,相关系数分别为-0.833、-0.906,柴胡根系的可溶性蛋白含量与越冬死苗率呈极显著负相关,相关系数为-0.949。结论:柴胡越冬期的游离脯氨酸、可溶性蛋白含量与越冬死苗率显著负相关,HB-sx与GS-dx的越冬死苗率较低,适宜在河北地区栽培。Objective:To study the correlation between the cold resistance and the contents of free proline,soluble protein,soluble sugar and other physiological indexes of six Bupleurum chinense germplasm,so as to provide theoretical basis for cold resistance evaluation,good seedlings screening and artificial cultivation of Bupleurum chinense germplasm.Methods:Six Bupleurum chinense seeds were collected from Dingxi of Gansu,Jinan of Shandong,Shiyan of Hubei,Shexian of Hebei,Wanrong of Shanxi and Longxi of Shaanxi,the serial numbers were GS-dx,SD-jn,HB-sy,HB-sx,SX-wr,SX-lx,respectively.The growth status and dead seedling rate of Bupleurum chinense were studied during the wintering and regreening stages.The free proline content was determined by ninhydrin colorimetry.The soluble protein content was determined by Coomassie Brilliant Blue G-250 staining method.The soluble sugar content was determined by anthrone colorimetry method.Results:The dead seedling rate in wintering stage of six Bupleurum chinense germplasms was significantly different.The dead seedling rate of HB-sx was 7.81%,followed by 12.87%of GS-dx.The dead seedling rate was SX-lx>SX-wr>HB-sy>SD-jn>GS-dx>HB-sx.During the wintering and regreening periods,the contents of free proline,soluble protein and soluble sugar in the roots and leaves of six Bupleurum chinense germplasms were significantly different.The free proline content of GS-dx roots in wintering period was 1.35 mg/g.The soluble protein content of HB-sx roots was 1.37μg/g and the soluble sugar content was 3.88%,and all contents were the highest.The free proline content of HB-sx leaves was 0.58 mg/g,the soluble protein content of GS-dx leaves was 0.97μg/g and soluble sugar content was 3.48%,and all contents were the highest.The free proline content in roots and leaves of Bupleurum chinense in wintering period was significantly negatively correlated with the dead seedlings rate in wintering stage,the correlation coefficients were-0.833 and-0.906,respectively.The soluble protein content of Bupleurum chinens
