摘 要:为了研究脂蛋白脂肪酶(lipoprotein lipase,LPL)基因在不同乳脂率德宏奶水牛乳腺组织中的表达情况,试验选取216头德宏奶水牛,采集乳样,用乳品分析仪测定乳脂率,德宏奶水牛的乳脂率大部分为(7.5±0.5)%,将其确定为中乳脂率组(M组),而乳脂率>8.0%的确定为高乳脂率组(H组),乳脂率<7.0%的确定为低乳脂率组(L组)。根据乳脂率测定结果,选择同一养殖小区中饲养条件及胎次相同、年龄相近、处于泌乳盛期的不同乳脂率的健康德宏奶水牛各3头,采集乳样,用乳成分分析仪测定乳脂率,采用气相色谱法测定脂肪酸含量;采集乳腺组织,采用实时荧光定量PCR法检测LPL基因mRNA表达水平,采用蛋白免疫印迹法检测LPL蛋白表达水平,并与德宏奶水牛乳中的乳脂率和脂肪酸含量进行相关性分析。结果表明:H组乳脂率、总脂肪酸和长链脂肪酸含量均极显著高于M组和L组(P<0.01),M组显著高于L组(P<0.05);H组不饱和脂肪酸含量极显著高于L组(P<0.01),显著高于M组(P<0.05),而M组与L组间差异不显著(P>0.05)。LPL基因mRNA和蛋白相对表达量均为H组极显著高于L组(P<0.01),而H组与M组、M组与L组间均差异不显著(P>0.05)。乳脂率与乳中总脂肪酸、长链脂肪酸和不饱和脂肪酸含量间呈极显著正相关(P<0.01);LPL基因mRNA表达水平与乳脂率、总脂肪酸含量、长链脂肪酸含量均呈显著正相关(P<0.05),而与不饱和脂肪酸含量无显著相关性(P>0.05)。说明LPL基因在乳脂合成过程中对脂肪酸的摄取和转运具有重要的调控作用。In order to study the expression of lipoprotein lipase(LPL) gene in mammary tissues of Dehong dairy buffalo with different milk fat contents. 216 Dehong dairy buffalos were selected in this and milk samples were collected for the determination of milk fat rate with milk composition analyzer. The most milk fat content of Dehong dairy buffalo was(7.5±0.5)%, which was determined as the medium fat group(M group), the>8.0% milk fat content was indeed defined as the high fat group(H group), and lower than 7.0% milk fat content was indeed defined as the low fat group(L group). Three healthy Dehong dairy buffalos in each group with the same feeding conditions, number of fetuses, similar age were selected. All animals were sampled at peak of lactation. Milk samples were collected and milk fat contents were tested by milk composition analyzer, and fatty acid contents were measured by gas chromatographic. The mRNA and protein levels of LPL in mammary tissue were measured by real-time PCR and western-blot, respectively. The correlation between the LPL gene expression and milk fat, fatty acid content in buffalo milk were analyzed. The results showed that the milk fat contents, total fatty acid and long chain fatty acid contents were extremely significantly higher in group H compare to group M and group L(P<0.01), group M was significantly higher than group L(P<0.05). The content of unsaturated fatty acid in group H was extremely significantly higher than group L(P<0.01), and group H was significantly higher than that in group M(P<0.05), but there was no significant difference between group M and group L(P>0.05). The mRNA and protein expression level of LPL gene in group H were extremely significantly higher than group L(P<0.01), while there was no significant difference when compare group H and group M or group M and group L(P>0.05).
