NCOA1基因mRNA表达及多态性与黔北麻羊产羔性状关联性分析

摘  要:为了研究核受体辅激活蛋白1(NCOA1)对黔北麻羊繁殖性能的影响,试验采用DNA测序技术,利用SeqMan软件分析测序结果初步筛选SNP位点,根据筛选出的SNP位点应用RNA在线二级结构预测软件中的RNA secondary structure prediction、ProtScale、DNAStar软件中的EditSeq分析NCOA1基因突变位点的二级结构、理化性质、亲疏水性、结构域,运用SPSS 20.0软件分析NCOA1基因型与产羔数的关联性;再应用实时荧光定量PCR方法检测NCOA1基因在黔北麻羊子宫、输卵管、垂体、下丘脑、卵巢5个器官中mRNA的表达水平。结果表明:在外显子7处发现2个SNP位点,命名为G144A和G155A,其中G144A属于同义突变,编码的氨基酸未发生改变,G155A属于错义突变,编码的精氨酸突变为赖氨酸,导致外显子7所编码的氨基酸自由能升高,稳定性降低,分子质量变大,等电点升高,疏水性增强,但未发现保守结构域和特殊结构域,即NCOA1基因外显子7编码的蛋白无特异结构和独立功能;对G155A位点进行基因分型,发现GG基因型个体产羔数分别比GA、AA基因型个体多0.249只(P>0.05)和0.377只(P<0.05);NOCA1基因mRNA在黔北麻羊5个器官中均有表达,在下丘脑中的表达量最高,在输卵管中的表达量最低,且卵巢、下丘脑中的表达量极显著高于输卵管、子宫(P<0.01),垂体中的表达量显著高于子宫和输卵管(P<0.05)。说明NCOA1基因对黔北麻羊繁殖性能有一定影响,有G155A突变位点的GG基因型也对黔北麻羊高产有一定影响,可作为筛选黔北麻羊高繁殖力品系的一个潜在指标。In order to study the effect of nuclear receptor coactivator protein 1(NCOA1) on the reproductive performance of Qianbei Ma goats, blood samples from lambs of Qianbei Ma goats were used to DNA sequencing of NCOA1 gene. The sequencing results were analyzed by SeqMan software SNP sites were screened. Then the secondary structure, physical and chemical properties, affinity/hydrophobicity and domain of the NCOA1 gene mutation were analyzed by RNA secondary structure prediction in RNA online secondary structure prediction software, ProtScale and EditSeq in DNA Star software based on the SNP sites. The association between NCOA1 genotype and lamb traits was analyzed by SPSS 20.0 software. The mRNA expression level of the NCOA1 gene in the uterus, fallopian tubes, pituitary, hypothalamus and ovary of Qianbei Ma goats was detected by quantitative real-time PCR. The results showed that two SNP sites were found at exon 7, named G144 A and G155 A. Among them, G144 A was a synonymous mutation, the encoded amino acid was not changed, G155 A was a missense mutation, and the encoded arginine mutation was lysine Acid, which lead to the increase of free energy of the amino acid encoded by exon 7, the decrease of stability, the increase of molecular mass, the increase of isoelectric point, and the increase of hydrophobicity. No conserved domain or special domain was found, that is, the protein encoded by exon 7 of NCOA1 gene has no specific structure and independent function. Genotyping of G155 A loci revealed that the number of lambs produced by GG genotype was 0.249 more(P>0.05) and 0.377(P<0.05);The mRNA of NOCA1 gene was expressed in all five organs of Qianbei Ma goats, the expression level in the hypothalamus was the highest, the expression level in the fallopian tube was the lowest, the expression level in the ovary and hypothalamus was significantly higher than that in the fallopian tube and uterus(P<0.01), and the expression in the pituitaries was significantly higher than that in the uterus and fallopian tubes(P<0.05).
