摘 要:为探讨丝裂原活化蛋白激酶p38(p38MAPK)抑制剂SB203580对热应激雄性雏鸡主要生殖激素含量的影响,试验将72只1日龄健康雄性文昌鸡雏鸡常规饲养2周,随机分为对照(CK)组、热应激(HS)组及热应激+SB203580(HS-I)组,每日中午将HS组与HS-I组雏鸡置于温度为(40.0±0.5)℃,相对湿度为(70±5)%的大型人工气候箱中热处理2 h(12:00—14:00),第3,4,5,6周龄末、热应激前0.5 h对HS-I组雏鸡按体重腹腔注射SB203580 5 mg/kg,HS组及CK组腹腔注射等量生理盐水;然后将HS组及HS-I组雏鸡置于人工气候箱中进行热应激[温度为(40.0±0.5)℃,相对湿度为(70±5)%]2 h(12:00—14:00),CK组雏鸡同时置于没有加热的人工气候箱中2 h。热应激结束后,各组随机抽取6只雏鸡立即采集颈动脉血液,并用酶联免疫吸附法测定雏鸡血浆中促性腺激素释放激素(GnRH)、卵泡刺激素(FSH)、黄体生成素(LH)、睾酮(T)等激素含量,试验进行4周。结果表明:与CK组相比,HS和HS-I组的GnRH、LH、T含量在第3,6周显著下降(P<0.05),HS-I组的FSH含量在第6周显著降低(P<0.05);在第5周,HS-I组FSH和T含量显著高于HS组(P<0.05)。说明热应激能显著降低雏鸡生殖激素含量,而SB203580可提高热应激雄性雏鸡FSH和T含量。To investigate the effect of mitogen-activated protein kinase p38(p38 MAPK) inhibitor SB203580 on the secretion of major reproductive hormones in heat-stressed male chicks, seventy two healthy 1-day-old male Wenchang chicks reared routinely for two weeks. All chicks were randomly divided into control(CK) group, heat stress(HS) group and heat stress + SB203580(HS-I) group. The chickens of the HS group and the HS-I group were placed in a large artificial climate chamber with temperature of(40.0±0.5) ℃ and relative humidity of 70%±5% for 2 h(12:00 — 14:00) at noon every day. From the end of age 3rd, 4th, 5th, and 6th weeks and 0.5 h before heat stress, the HS-I group were intraperitoneal injected SB203580(5 mg/kg) according to their body weight, and the HS group and CK group chicks were intraperitoneal injected with the same amount of normal saline. Then, HS group and HS-I group chicks were placed in the artificial climate box for heat stress [temperature:(40.0±0.5) ℃, relative humidity: 70%±5%] for 2 h(12:00—14:00), and CK group chicks were placed in the artificial climate box without heating for 2 h. After heat stress, 6 chicks in each group were randomly selected to collect carotid blood immediately, and the concentrations of gonadotropin releasing hormone(GnRH), chicken follicle-stimulating hormone(FSH), chicken luteinizing hormone(LH), chicken testosterone(T) and other hormones in plasma were determined by enzyme-linked immunosorbent assay. The experiment was conducted for 4 weeks. The results showed that compared with the CK group, the contents of GnRH, LH, and T in the HS and HS-I groups decreased significantly at the 3rd and 6th weeks(P<0.05), while the FSH contents in the HS-I group decreased significantly at the 6 th week(P<0.05);At week 5, the contents of FSH and T in HS-I group was significantly higher than those in HS group(P<0.05).
