摘  要:目的观察川续断皂苷Ⅵ经ATF6通路对H9c2心肌细胞氧化应激和凋亡的保护作用。方法2019年6—9月于邯郸市第一医院实验室进行实验。应用不同条件对H9c2细胞培养,对照组加入无血清RPMI-1640培养基,H2O2组加入终浓度为200μmol/L的H2O2,川续断皂苷Ⅵ低、中、高剂量组分别加入终浓度为25、50、100μmol/L的川续断皂苷Ⅵ,同时给予终浓度为200μmol/L的H2O2,继续培养24 h后检测H9c2细胞增殖和凋亡情况,同时检测H9c2细胞内ROS、LDH、MDA、Bcl-2、Bax、Caspase-3、ATF6、GRP78和PDIA6水平。结果与对照组比较,H2O2组H9c2细胞凋亡率、ROS、LDH、MDA、Bax和Caspase-3水平升高(P<0.05),细胞增殖率、Bcl-2、ATF6、GRP78和PDIA6水平降低(P<0.05);与H2O2组比较,各川续断皂苷Ⅵ组H9c2细胞凋亡率、ROS、LDH、MDA、Bax和Caspase-3水平降低(F/P=23.957/0.000、132.452/0.000、85.716/0.000、30.485/0.000、15.156/0.023、15.022/0.025),细胞增殖率、Bcl-2、ATF6、GRP78和PDIA6水平升高(F/P=41.542/0.000、23.118/0.000、25.416/0.000、9.240/0.037、20.276/0.000),且呈剂量依赖性(P<0.05)。结论川续断皂苷Ⅵ通过激活ATF6通路,减轻H2O2诱导的H9c2细胞氧化应激和抑制细胞凋亡,从而发挥心肌保护作用。Objective To observe the protective effects of Dipsacus asper saponin VI on oxidative stress and apoptosis of H9c2 cardiomyocytes via ATF6 pathway.Methods The experiment was carried out in the Laboratory of Handan First Hospital from June to September 2019.H9c2 cells were cultured under different conditions.The control group was added with serum-free RPMI-1640 medium.In H2O2 group,H2O2 with final concentration of 200μmol/L was added.In the low,medium and high dose groups,the final concentration of Dipsacus asper saponinⅥwas 25,50 and 100μmol/L,and the final concentration of H2O2 was 200μmol/L.The proliferation and apoptosis of H9c2 cells were detected after 24 hours.The levels of ROS,LDH,MDA,BCL 2,Bax,caspase 3,ATF6,GRP78 and pdia6 in H9c2 cells were detected.Results Compared with the control group,the apoptosis rate,ROS,LDH,MDA,Bax and caspase 3 levels of H9c2 cells in H2O2 group were increased(P<0.05),while cell proliferation rate,Bcl-2,ATF6,GRP78 and pdia6 levels were decreased(P<0.05).Compared with H2O2 group,H9c2 cell apoptosis rate,ROS,LDH,MDA,Bax and caspase 3 levels were decreased(F/P=23.957/0.000,132.452/0.000,85.716/0.000,30.485/0.000,15.156/0.023,15.022/0.025),cell proliferation rate and Bcl-2 level were significantly decreased in all groups 2.The levels of ATF6,GRP78 and pdia6 increased in a dose-dependent manner(F/P=41.542/0.000,23.118/0.000,25.416/0.000,9.240/0.037,20.276/0.000)in a dose-dependent manner(P<0.05).Conclusion Dipsacus asper saponin VI can protect H9c2 cells from oxidative stress and apoptosis induced by H2O2 by activating ATF6 pathway.
