摘 要:青霉素G酰化酶(penicillin G acylase,PGA)是生物催化合成青霉素类和头孢菌素类等β-内酰胺抗生素的重要工业用酶,在药物中间体的合成、手性化合物的拆分、多肽合成过程中基团的保护和去保护等领域有重要的应用.设计与选择PGA固定化适宜的载体和酶催化反应介质,是提高PGA的合成活性和稳定性的有效途径.综述离子液体在青霉素G酰化酶固定化过程中的溶剂化作用和对载体的修饰作用以及催化反应的介质效应,分析离子液体阴阳离子的结构特点和体积大小、溶剂微环境、水活度及亲疏水性等因素对PGA催化性能的影响;总结介质效应及离子液体功能化提高催化剂活性的作用机制,并对离子液体在酶催化技术的应用前景提出展望.Penicillin G acylase(PGA)is an important industrial enzyme for biocatalytic synthesis ofβ-lactam antibiotics such as penicillins and cephalosporins,which has an important value for the application to the fields such as the pharmaceutical intermediates synthesis,the chiral compounds resolution,and protection and deprotection of groups in the process of polypeptide synthesis.It is an effective way to improve the synthetic activity and stability of PGA to design and select carrier and enzyme catalytic reaction medium suitable for PGA immobilization.The article reviewed the solvation effect of ionic liquids in the immobilization of penicillin G acylase,its modification function on carriers,the medium effect of catalytic reaction,and analyzed the influence of structure,size and volume,solvent microenvironment,water activity and hydrophobicity of ionic liquid anions and cations on the catalytic performance of PGA;it summarized the mechanism of medium effect and ionic liquid functionalization improving catalyst activity,and prospected the application outlook of ionic liquid in enzyme catalysis technology.
