摘 要:目的探讨被动吸烟对雄性大鼠大脑组织中wnt/β-catenin信号通路的影响。方法雄性SD大鼠40只分为正常对照组(新鲜空气),低剂量组(10根香烟/d),中剂量组(20根香烟/d),高剂量组(30根香烟/d),每组10只动物。被动吸烟12周后,处死所有动物。苏木素-伊红(HE)染色法观察大脑皮质组织的病理结构改变,Western blot方法分析大脑组织中wnt-2b、GSK-3β、β-catenin和磷酸化β-catenin(P-β-catenin)蛋白水平的表达。结果被动吸烟12周后,雄性SD大鼠的大脑组织HE染色结果显示被动吸烟引起了大脑皮质结构分层紊乱,间质毛细血管增生伴扩张充血,随吸烟剂量的增加而持续加重。Western blot结果显示,wnt-2b蛋白表达水平在中、高剂量组出现明显的下调(P<0.05),β-catenin和P-β-catenin蛋白水平在高剂量组及GSK-3β蛋白在各剂量组均出现明显的上调,且差异均有统计学意义(P<0.05)。结论被动吸烟会引起雄性大鼠大脑组织结构紊乱,并且可通过降低wnt-2b、增加GSK-3B及P-β-catenin蛋白表达水平而抑制wnt/β-catenin信号通路。Objective To investigate the effects of passive smoking on the wnt/β-catenin signaling pathway in the brain tissue of male rats.Methods Forty male SD rats were divided into normal control group(fresh air),low dose group(10 cigarettes/day),medium dose group(20 cigarettes/day),and high dose group(30 cigarettes/day),10 animals per group.Cigarette smoke was passively inhaled after 12 weeks,all the animals were sacrificed.HE staining was used to determine the pathological changes of brain tissue.Western blot analysis was used to analyze the expressions of wnt-2b,β-catenin,phosphorylatedβ-catenin and GSK-3βin brain tissue.Results After 12 weeks of passive smoking,HE staining of brain tissue showed that passive smoking caused stratification disorder of cerebral cortex,interstitial capillary proliferation with dilated hyperemia,and this injury become heavier with increasing smoking dose.Western blot result showed that both wnt-2b and GSK-3βprotein level were significantly down-regulated in the middle and high dose group(P<0.05),a significant up-regulation ofβ-catenin and phosphorylatedβ-catenin protein appeared in high dose group,and the difference was statistically significant(P<0.05).Conclusion Passive smoking causes structural disorder in the brain of male rats,and can inhibit the wnt/β-catenin signaling pathway by decreasing wnt-2b,increasing the expression levels of GSK-3βand P-β-catenin.
