摘  要:目的探讨汽油添加剂甲基叔丁基醚(methyl tert-butyl ether,MTBE)对人DNA聚合酶β低表达细胞的DNA损伤情况。方法运用RNA干扰技术构建人支气管上皮细胞(HBE)DNA聚合酶β(polβ)低表达细胞株HBEpolβ-;常规培养HBE-polβ-、pEGFP-C1空载体细胞及HBE细胞至对数生长期,分别以终浓度为0、2、4、8和16μmol/L的MTBE染毒培养48 h;以单细胞凝胶电泳(SCGE)检测各组细胞的DNA损伤情况。结果HBE-polβ-细胞中polβ表达仅相当于正常HBE细胞的18.38%;当MTBE染毒浓度为2、4、8和16μmol/L时,各组细胞均出现明显的彗星拖尾现象,且HBE-polβ-组细胞DNA彗星Olive尾矩与对照组比较,差异具有统计学意义(P<0.05)。结论polβ的异常表达对MTBE所致DNA损伤可能易感。Objective To investigate the DNA damage induced by methyl tert-butyl ether(MTBE),a gasoline additive,in human bronchial epithelial cells with low level of DNA polymerase beta(HBE-polβ-).Methods RNA interference technique was used to construct HBE-polβ-cell line.In logarithmic growth phase,different groups of HBE-polβ-,pEGFP-C1 and HBEcells were cultured with different concentrations of MTBE(0,2,4,8 and 16μmol/L)for 48 h,respectively.Single cell gel electrophoresis(SCGE)analysis was used to detect DNA damage of each group.Results The expression level of polβin HBE-polβ-group was only about 18.38%of that in normal HBE cells.When MTBE concentration was at 2,4,8 and 16μmol/L,obvious comet tails were observed in all groups,and there were statistically significant difference in Olive tail moment between groups of HBE-polβ-and controls(P<0.05).Conclusion HBE cells with abnormal expression of polβmay be susceptible to DNA damage induced by MTBE.
