高效液相色谱法评估左氧氟沙星的眼内通透性

摘　　要：目的:评估左氧氟沙星滴眼液入眼通透性。采用高效液相色谱法(HPLC)测定33例白内障手术患者房水中左氧氟沙星滴眼液的浓度。方法:共33例接受超声乳化术的患者,术前、术后3d均使用左氧氟沙星点眼,每6h滴1滴,术前1h停止给药。采用HPLC和荧光检测器测定左氧氟沙星在房水中的浓度。结果:采用HPLC测定房水中左氧氟沙星含量,验证了该方法简便、有效、灵敏。左氧氟沙星浓度在1.95×10-3-1.50μg/mL的范围内呈线性关系,在房水中平均含量为0.3399±0.03405μg/mL。结论:本研究结果显示,白内障术前常规剂量(每6h滴1滴)0.5%左氧氟沙星不会引发急性细菌性眼内炎。AIM:To evaluate the levofloxacin eye drop into human eye penetration,levofloxacin eye drop concentrations in human ocular aqueous of 33 patients undergoing cataract surgery were measured by high performance liquid chromatography(HPLC).METHODS:Totally 33 volunteer patients who scheduled for phacoemulsification surgery received one drop of levofloxacin every 6h for 3d before and on the day of surgery,administration of drug was stopped 1h before surgery.Levofloxacin concentration in aqueous humor was measured by HPLC method with fluorescence detector.RESULTS:A simple,effective and sensitive HPLC method for determination of levofloxacin in human ocular aqueous was validated.Linearity was shown for levofloxacin concentration over a wide range of 1.95×10-3-1.50μg/mL.The mean aqueous level of levofloxacin was 0.3399±0.03405μg/mL.CONCLUSION:Results from the present study demonstrate that topical administration of levofloxacin 0.5%before cataract surgery with routine dose(one drop every 6h)unable to reach MIC90 for most common microorganism causing acute bacterial endophthalmitis.
