泉州中华蜜蜂遗传多样性分析

摘　　要：采用33个形态标记、38个微卫星标记和tRNAleu^COII片段的线粒体标记,对泉州7个县市,共487只中华蜜蜂进行遗传分化、遗传特征和遗传多样性分析.研究结果表明,泉州样点均属于同一种群,无遗传分化.线粒体tRNAleu^COII片段共检测到29种单倍型,其中9种为新发现单倍型;泉州中华蜜蜂处于中度多样性水平,期望杂合度(He)在0.4407~0.5131之间,观察杂合度(Ho)在0.4062~0.5124之间,平均多态信息含量(PIC)在0.4091~0.4765之间,单倍型多样性(Hd)的范围0.241~0.739,平均核苷酸差异数(K)为0.339~0.926,核苷酸多样度(Pi)为0.00096~0.00334.To study the genetic diversity and geographical differentiation of Apis cerana habitating in Quanzhou City,southeast China's Fujian Province.497 samples of A.cerana from 7 counties were analyzed using 33 morphological markers,38 microsatellite markers and mitochondrial marker of tRNAleu^COII fragments.The results showed that A.cerana from Quanzhou do not genetically differentiate and all belong to 1 population.A total of 29 haplotypes were detected based on tRNAleu^COII fragments,of which 9 were newly discovered haplotypes.This honeybee population stays in moderate diversity level,with the expected heterozygosity(He)ranging from 0.4407 to 0.5131 and heterozygosity observed(Ho)ranging from 0.4062 to 0.5124.Polymorphisms information content averaged between 0.4091 and 0.4765,and haplotype diversity(Hd)varied between 0.241 and 0.739.Average nucleotide difference(K)ranged from 0.339 to 0.926,resulting in a nucleotide diversity of 0.00096 to 0.00334.
