摘 要:【目的】探讨SSR分子标记辅助选择技术对不同遗传背景甜糯双隐性玉米种质的鉴定效果,为甜糯双隐性玉米自交系的创制提供技术支撑。【方法】利用与糯质基因(waxy)紧密连锁的SSR标记引物phi022、phi027、phi061分别对常规甜、糯玉米杂交组配及甜、糯玉米染色体杂交手段获得的共354份材料进行了甜糯双隐基因型鉴定。【结果】标记引物phi022可作为常规甜、糯玉米杂交F1在甜质背景下选育的84份材料和F1在糯质背景下选育的186份材料糯质基因的前景选择标记,鉴定出甜糯双隐性玉米纯合体的比例分别为19.05%和100.00%。标记引物phi061可作为甜、糯玉米染色体杂交手段获得的84份材料糯质基因的前景选择标记,鉴定出甜糯双隐性玉米纯合体的比例为46.43%。【结论】SSR分子标记辅助选择技术鉴定出的甜糯双隐性玉米纯合体的比例与理论值大致吻合,可作为苗期快速选择甜糯双隐性玉米材料的技术手段。值得注意的是,材料遗传背景、标记引物特异性会影响SSR分子标记鉴定效果,可根据育种需要结合传统鉴定方法来提高甜糯双隐性玉米自交系的选择效率和创制准确性。【Objective】The present paper aimed to discuss the effect of SSR molecular marker-assisted selection technology on the identification of double recessive sweet-waxy maize germplasm with different genetic backgrounds,and to provide technical support for the creation of sweet-waxy double recessive maize inbred lines.【Method】The SSR marker primers,viz.,phi022,phi027,phi061,closely linked with the waxy gene(waxy),were used to identify the sweet-waxy double recessive genotypes of 354 materials obtained by conventional sweet and waxy maize hybridization and chromosome hybridization.【Result】The marker primer phi022 could be used as the foreground selection marker to select the 84 sweet materials of conventional sweet and waxy maize hybrid F1 under sweet background and the 186 materials of conventional sweet and waxy maize hybrid F1 under waxy background.The percentage of homozygotes of double recessive sweet-waxy maize was 19.05%and 100.00%respectively.The marker primer phi061 could be used as the foreground selection marker to select the 84 materials obtained by chromosome hybridization.The percentage of homozygotes of double recessive sweet-waxy maize was 46.43%.【Conclusion】The proportion of double recessive sweet-waxy maize homozygote identified by SSR molecular marker-assisted selection technique roughly agreed with the theoretical value.It could be used as a rapid technical method to select double recessive sweet-waxy maize at seedling stage.It should be noted that the genetic background of the material and the specificity of the marker primer would affect the identification effect of SSR molecular markers.The selection efficiency and creation accuracy of double recessive sweet-waxy maize inbred lines could be improved according to breeding needs combined with traditional identification methods.
